摘要
目的探讨重组成骨蛋白-1(r OP-1)对钛-6铝-4钒微粒(Ti-6Al-4V)诱导成骨细胞中炎性因子如肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)和白介素-6(IL-6)的影响。方法将成骨细胞分为正常对照组、r OP-1组、Ti-6Al-4V组,Ti-6Al-4V+r OP-1组,采用Elisa法检测各组细胞培养72 h后TNF-α、IL-1β和IL-6含量的变化。结果培养72 h后,与Ti-6Al-4V组比较,Ti-6Al-4V+r OP-1组TNF-α、IL-1β和IL-6含量下降,抑制率分别为25.67%、7.78%和21.86%,差异有统计学意义(均P<0.05)。结论 r OP-1可抑制Ti-6Al-4V诱导成骨细胞炎性因子TNF-α、IL-1β和IL-6,为为防治关节假体无菌性松动提供新的方法。
Objective To investigated recombinant osteogenic protein 1 affects of TNF-α, IL-1β, and IL-6 on Ti-6Al-4V-induced osteoblast.Methods Osteoblasts were divided into 4 groups: control group, rOP-1 group, Ti-6Al-4V group and Ti-6Al-4V with rOP-1 group, Elisa was used to assay effects of inflammatory cytokines after 72 h incubation with Ti-6Al-4V (rOP-1).Results Compared with the osteoblasts with particle group, after 72 h-incubation, the TNF-α, IL-1β, and IL-6 of osteoblasts in Ti-6Al-4V with rOP-1 group was significantly decreased , inhibition 25.67%, 7.78%, and 21.86%, respectively (P〈0.05).Conclusion rOP-1 decreased level of TNF-α, IL-1βand IL-6 on osteoblast and could be therapeutic candidate for the prevention of artificial joint aseptic loosening .
