摘要
目的探讨海带多糖对人肝癌bel-7402细胞存活和凋亡的影响。方法 1体外培养人肝癌bel-7402细胞,加入海带多糖(浓度分别为5、10、20、30 g/L)继续培养48 h,另将不用药物干预的肿瘤细胞组设为对照组;倒置显微镜观察细胞形态的变化,MTT法测定细胞存活率。2体外培养人肝癌bel-7402细胞,设对照组及海带多糖组,海带多糖用药组分别加入海带多糖(10、20、30 g/L),采取相应干预措施。用流式细胞仪检测细胞周期和细胞凋亡情况。结果 1海带多糖(5、10、20、30 g/L)分别作用bel-7402细胞48 h后,随着海带多糖浓度的增加,bel-7402细胞逐渐变小、固缩、变形;细胞存活率明显降低。2海带多糖作用48 h后,bel-7402细胞凋亡率为(14.8±1.7)%,坏死率为(19.8±2.3)%,与对照组比较,差异有统计学意义(P<0.05)。结论海带多糖可抑制bel-7402细胞存活,促进bel-7402细胞凋亡及坏死。
Objective To observe the effect of laminarin on survival and apoptosis of human hepatocarcinoma cell of Bel-7402. Methods 1 Cultured human hepatocarcinoma cell of Bel-7402 in vitro,after added with the 5,10,20,30,40 g / L laminarin,cultured for 48 h. Without adding laminarin as the control group,we observed the changes of cells by inverted microscope and detected the cell viability by MTT assay. 2 Cultured human hepatocarcinoma cell of Bel-7402 in vitro,three groups： The control group,laminarin group and laminarin ＋ medicine group were treated with 10,20 and 30 g / L respectively. The cell cycle and apoptosis rate were analyzed by flow cytometer. Results 1 After bel-7402 cells were cultured using 5,10,20,30,40 g / L laminarin for 48 h,with the increasement of laminarin concentration,bel-7402 cells were shrinked and changes. The cell survival rate was significantly decreased. 2 After 48 h,the bel-7402 cells apoptosis rate was（ 14. 8 ± 1. 7） % and death rate was（ 19. 8 ± 2. 3）,with significant difference from the control group（ P〈0. 05）. Conclusion Laminarin can inhibit survival rate,and promote the apoptosis and necrocytosis of bel-7402 cells.
