摘要
目的:观察益视汤对脾气虚型兔RD自动复位后细胞凋亡及机体氧化应激状态的影响。方法:实验兔按随机数字表法分为正常对照组(A)、RD自动复位组(B)、脾气虚型RD自动复位组(C)及益视汤治疗组(D),分别于RD自动复位模型制备后10d、20 d、30 d随机处死取材,采用透射电镜及TUNEL染色观察视网膜各层细胞凋亡的发生,取血清行SOD、MDA测定。结果:电镜下可见B、C、D组视网膜内外核层出现凋亡特征性形态改变细胞及TUNEL染色阳性细胞,比较细胞凋亡指数,C组较B组明显升高;D组较C组显著下降。与A、B组比较,C组血清SOD活性明显降低,MDA值明显升高,D组较C组SOD活性显著升高,MDA降低。结论:脾气虚证加重机体氧化损伤及RD自动复位后细胞凋亡的发生,使用益视汤能够提高机体的抗氧化能力,降低细胞凋亡的发生。
Objective: To investigate the influence of Chinese medicinal decoction with tonifying qi and yin and diuresis on retinal apoptosis in reattached retina of deficiency of spleen-qi rabbit model. Method: 42 pigment rabbits were randomly divided into normal control group( A),RD automatically reattachment group( B),and RD automatically reattachment with deficiency of spleen-qi group( C) and Chinese medicinal decoction treatment group( D). Then the reattachment retinal apoptosis were detected with TUNEL staining and electron microscopy in postoperative 10 d,20 d,and30 d. SOD activities and MDA levels of blood serum were also checked. Result: The morphological features of apoptosis,such as nuclear chromatin condensation and fragmentation as well as cell shrinkage presented in the ONL and INL of B,C and D groups. The results of TUNEL staining revealed the positive cells presented in not only ONL but also INL and NFL in the reattached retina,and apoptotic indexes of C groups increased obviously in 10 d post operation. Compared with A and B groups,the activities of serum SOD decreased and the content of MDA increased. The Chinese medicinal decoction could obviously inhibit the occurrence of retinal apoptosis at any time points,enhanced serum SOD activities and reduced the levels of serum MDA. Conclusion: Our results suggest that the pathology spleen-qi syndrome increase oxidative stress damage and aggravate the retinal apoptosis in retina reattachment. The Chinese medicinal decoction with tonifying qi and yin and diuresis is a potent neuroprotective agent that can salvage retinal neurons in rabbits with retinal reattachment and elevate the ability of antioxidation.
