hsa-miR-136和PPP2R2A在骨巨细胞瘤组织中的表达及其意义

摘要 目的:检测hsa-miR-136和PPP2R2A在骨巨细胞瘤组织中含量,并预测二者的作用位点。方法:采用Real-time PCR方法检测hsa-miR-136和Western-blot方法检测PPP2R2A在骨巨细胞瘤组织和瘤旁组织含量。利用miRNA靶基因预测软件Target Scan,对hsa-miR-136的靶基因进行预测,Bibi Serv软件分析hsa-miR-136与靶基因结合的自由能。结果:hsa-miR-136在骨巨细胞瘤组织中的含量明显高于瘤旁组织(P<0.05)。PPP2R2A在骨巨细胞瘤组织中的含量明显低于瘤旁组织(P<0.05)。hsa-miR-136和PPP2R2A与骨巨细胞瘤患者的病理特征没有明显相关性。Kaplan-Meier生存曲线分析显示hsa-miR-136和PPP2R2A与骨巨细胞瘤患者生存时间具有显著相关性(P<0.05)。结论:hsa-miR-136和PPP2R2A在骨巨细胞瘤组织中表达异常,提示hsa-miR-136和PPP2R2A可能与骨巨细胞瘤的发生、发展有密切关系。 Objective:To determine the levels of hsa - miR - 136 and PPP2R2A in giant cell tumor of bone. And binding sites of hsa - miR - 136 and PPP2R2A were predicated. Methods:The levels of hsa - miR - 136 and PPP2R2A in giant cell tumor of bone were detected by using Real - time PCR and Western - blot,respectively. The candidate target genes of hsa - miR - 136 were predicated by using TargetScan software. The minimum free energy predicted for hybridization was determined by BibiServ analysis. Results:The levels of hsa - miR - 136 were higher in cancer tissues than that in normal tissues(P 〈 0. 05). The levels of PPP2R2A were lower in cancer tissues than that in normal tissues(P 〈 0. 05). The levels of hsa - miR - 136 and PPP2R2A were not related with the pathological fea-ture of the patients with giant cell tumor of bone. Both hsa - miR - 136 and PPP2R2A were associated with the surviv-al rate of the patients with giant cell tumor of bone by using Kaplan - Meier survival curve analysis(P 〈 0. 05). Con-clusion:Abnormal hsa - miR - 136 and PPP2R2A were observed in giant cell tumor of bone cancer tissues,which suggest that there was association between hsa - miR - 136 and PPP2R2A and the development of giant cell tumor of bone.
