黄芪注射液体外促进宫颈癌细胞增殖及其调控分子表达分析 被引量:2

The expression analyse of regulators on promoted proliferation of cervical cancer cells in vitro by Injection of Astragalus Mongholicus

摘要 目的:分析黄芪注射液对宫颈癌细胞的促增殖效应及其相关调控分子表达的影响。方法:将人宫颈癌Hela细胞与系列稀释后终浓度分别为生药7.82 g/L、15.63g/L、31.25g/L、62.50g/L、125g/L、250g/L、500g/L、1000g/L、2000g/L、4000g/L(相当于每升原药材克数)的黄芪注射液共同孵育,噻唑蓝(MTT)法检测细胞生长增殖的A492值,流式细胞计数(FCM)分析细胞周期各时相细胞百分率及凋亡指数(AI)、增殖指数(PI);免疫细胞化学法检测细胞中PCNA、Cyclin A、Bcl-2、Bax等增殖凋亡相关调控分子表达的变化,计算表达阳性细胞百分率,并以图像分析系统定量分析阳性表达强度。结果:生药125g/L^4000g/L的黄芪注射液作用后,可使Hela细胞生长增殖的A492值呈递进性显著增高;Hela细胞G0/G1期比例及AI值显著降低,G2/M期比例及PI值显著升高,S期比例未见显著差异。同时,Hela细胞中PCNA阳性表达率、Cyclin A及Bcl-2阳性表达率及表达强度明显增高,Bax的阳性表达率及表达强度明显降低。结论:黄芪注射液可通过影响Hela细胞增殖凋亡相关调控分子的表达而发挥促进生长增殖的作用。黄芪制剂应用于宫颈癌患者及具此类肿瘤诱发体质人群的其他疾病治疗中须慎用。 Objective: To analyze the promoting effect of Astragalus mongholicus Bge (AMB) on the proliferation of cervical cancer cells and ex- pressions of their regulators. Methods: Hela cells ( human cervical cancer cells) were co-incubated with AMB injection at different final con- centrations which equals to the gram of original medicinal materials (7.82 g/L, 15.63g/L, 31.25g/L, 62.50g/L, 125g/L, 250g,/L, 500g/ L, 1000g/L, 2000g/L and 4000g/L, respectively). The proliferation of Hela cells was observed, values of A492 were measured by MTI' method. Their cell cycles, apoptosis index (AI) and proliferation index (PI) were calculated through flow cytometry (FCM). Expressions of PCNA, Cyclin A, Bcl-2 and Bax in Hela cells were analyzed through immunocytochemical staining, percentages of positive cells were calcu- lated, and the intensity of positive staining was measured by image analysis system synchronously. Results: AMB accelerated the growth and proliferation of Hela cells along with the increased concentrations ranged from original medicinal materials 125g/L to 4000g/L ( all P 〈 0. 05 ). And the proportion of cells in stage of G0/G1 and values of AI decreased, that in stage of G2/M and values of PI elevated greatly ( all P 〈 0.05), but no significant changes in stage of S were shown ( P 〉 0.05). Meanwhile, after administered with AMB, the positive percent- age of PCNA and both the percentage and the intensity of Cyclin A and Bcl-2 expressed in Hela cells were higher, and those of Bax were low- er significantly ( all P 〈 0.05 ). Conclusion : AMB injection can exert the promoting effect on the proliferation of Hela cells via the influence on expressions of regulators. AMB should be used cautiously and thoughtfully for the patients with cervical cancer and certain group that is sensitive for this kind of tumor.
