摘要 目的:本课题组之前的实验结果提示纳米银具有强有力的杀菌作用,本实验将探讨纳米银的体外生物安全性。方法:将人牙龈成纤维细胞与0.1%、0.5%、1.0%、1.5%和2%的纳米银-PLGA载体共同培养一定的时间,利用MTT法检测人牙龈成纤维细胞的增殖情况,并在骨向诱导培养基条件下分析人牙龈成纤维细胞碱性磷酸酶活性。结果:与对照组相比,人牙龈成纤维细胞的增殖情况在不同浓度的纳米银-PLGA组未出现明显减少,各组间也无明显差异。在骨向诱导培养条件下,人牙龈成纤维细胞表现出一定碱性磷酸酶活性,并且该活性未收到各浓度纳米银的影响。结论:本研究所采用的20~40nm银对人牙龈成纤维细胞具有良好的体外生物安全性。 Objective: To analyze the in vitro biosafety of 20--40nm nanosilver. Methods: Different concentration of nanosilver and human gingival fibroblast cell was cultured for 2,4 and 6 day,the cell proliferation was determined by MTT. The ALP activity of human gingival fibroblast cell under differentiated medium was detected at day 9,12 and 15. Results: The proliferation of human gingival fibroblast cell was similar in different group. And the ALP activity of human gingival fibroblast cell was not affected by 20- 40nm nanosilver. Conclusion: The 20- 40nm nanosilver had perfect hiosafety to human gingival fihroblast cell in vitro.
