响应面法优化超声波提取黑葵花籽皮花色苷工艺 被引量:2

Optimization of Ultrasonic Wave Extraction of Anthocyanins from Black Sunflower Seed Shell by Response Surface Methodology

摘要 以黑葵花籽皮为研究对象,以花色苷提取液的OD530nm值作为提取效果指标,采用超声波法对黑葵花籽皮花色苷的提取工艺进行了研究。考察提取溶剂的乙醇体积分数、超声功率、液料比对提取液OD530nm值的影响。在单因素试验的基础上,采用三因素三水平的响应面分析法对黑葵花籽皮花色苷的提取工艺进行了优化,依据回归分析确定最优提取条件。结果表明:最佳提取工艺条件为采用0.5%盐酸-71.5%乙醇作为提取溶剂,超声功率为390 W,液料比为19∶1(mL/g)。在此工艺条件下,花色苷提取液OD530nm值达最大,为0.878。 Taking black sunflower seed shells as the research object,the OD530 nmvalue of anthocyanin extracting solution is used as measure indicator,ultrasonic wave is applied for extracting anthocyanins from black sunflower seed shell. The effects of ethanol concentration,ultrasonic power and liquid-solid ratio on the OD530 nm value of extracting solution are investigated.On the basis of single factor investigation,response surface methodology with three factors at three levels is adopted to optimize the extraction technology of anthocyanins from black sunflower seed.The best extraction parameters are determined by the regression analysis.The results show that the optimum conditions of extraction are as follows:0.5% hydrochloric acid-71.5% ethanol as extracting solvent,ultrasonic power of 390 W,liquid-solid ratio of 19∶1(mL/g).Under the optimized conditions,OD530 nmvalue of anthocyanin extracting solution reaches a maximum of 0.878.
