Fli-1在外周血T/B细胞中的表达及其与系统性红斑狼疮的相关性研究 被引量:4

Expression of Fli-1 in circulating T /B cells and their correlations with systemic lupus erythematosus

摘要 目的检测转录因子Fli-1蛋白在外周血T细胞与B细胞上的表达水平,分析其表达水平与系统性红斑狼疮(systemic lupus erythematosus,SLE)患者疾病活动度、实验室指标及临床表现之间的关系。方法选取来自安徽医科大学附属省立医院的28例SLE患者作为病例组,选择25例健康志愿者作为正常对照。利用流式细胞仪(flow cytometry,FCM)检测外周血单核细胞(peripheral blood mononuclear cells,PBMCs)T细胞和B细胞中Fli-1蛋白表达情况。结果 SLE患者外周血T细胞上Fli-1平均荧光强度(mean fluorescence intensity,MFI)高于对照组,差异有统计学意义(t=3.28,P=0.003),而SLE病例与对照组间,Fli-1在外周血B细胞上的表达差异无统计学意义(Z=-0.87,P=0.383)。SLE病人外周血T/B细胞上Fli-1的表达水平与糖皮质激素的平均用量间均呈正相关(均有P<0.05),但并未发现Fli-1在T细胞和B细胞上的表达水平与SLE疾病活动指数间的关联。对多项临床实验室检测指标与Fli-1的关联分析结果显示,低补体的SLE患者,其Fli-1在外周血T细胞与B细胞上的表达水平均高于无低补体表现的SLE患者(均有P<0.05)。结论 SLE病人外周血T细胞上Fli-1表达水平增加,提示Fli-1可能参与了SLE的发病,其具体作用机制仍有待进一步研究。 Objective To compare the expressions of Fli-1 in the T/B cells between the systemic lupus erythematosus( SLE) patients and healthy controls and analyze the relationships between the expressions of Fli-1 in the T /B cells and SLE disease activity,clinical,and laboratory data. Methods 28 SLE patients were recruited from Anhui Provincial Hospital. 25 normal healthy volunteers were also recruited as controls. The FCM system was used to detect the expressions of Fli-1 in T cells and B cells. Results The mean fluorescence intensity( MFI) of Fli-1 in T cells increased in the PBMCs of patients with SLE compared with healthy controls( t = 3. 28,P = 0. 003). However,no significant differences were found in the MFI of Fli-1 in B cells( Z =- 0. 87,P = 0. 383). There was a positive correlation between the expression level of Fli-1 in peripheral blood T /B cells and dose of glucocorticoids( both P 0. 05). But no significant association was found between Fli-1 expression levels and SLEDAI score. Fli-1 expression levels on T / B cells of SLE patients with low complement was significantly higher than that of the patients without low complement. Conclusions Increased Fli-1 expression levels in the peripheral blood T cells of SLE patient,indicate that Fli-1 may be involved in the pathogenesis of the disease. The specific mechanism remains to be further explored.
