葡萄糖转运蛋白-1在糖尿病早期大鼠视网膜的表达

摘要 目的通过检测糖尿病早期大鼠视网膜葡萄糖转运蛋白-1(glucose transporter-1,GLUT1)表达的变化,揭示GLUT1对早期糖尿病视网膜的影响。方法选取鼠龄为12周的健康雄性Wistar大鼠80只,随机选取40只采用链脲佐菌素左下腹腔一次性注射建立1型糖尿病模型(实验中4只因严重感染而死),另取40只作为对照组注射同等量柠檬酸缓冲液。模型组大鼠建模成功后分别在4周、8周、12周、16周(每周9只大鼠)测定血糖和糖化血红蛋白,取同时期对照组(每周10只)大鼠做相同检测。免疫组织化学方法检测视网膜GLUT1蛋白表达,利用ADP酶组织化学染色方法进行视网膜铺片,了解视网膜血管改变情况。结果糖尿病组血糖及糖化血红蛋白显著高于对照组。免疫组织化学染色、光镜形态学观察显示糖尿病组16周视网膜节细胞层细胞排列极其紊乱,但未见出血,视网膜内核层水肿,内外核层细胞排列紊乱。糖尿病组GLUT1表达较对照组大鼠视网膜中阳性染色颗粒均明显增加。糖尿病组视网膜GLUT1表达的平均积分光密度在4周(63.903 3±42.245 3)、8周(96.369 1±18.697 2)、12周(102.002 8±23.998 9)、16周(130.408 7±72.745 1)依次有所增强,其中8周和12周差异无统计学意义,其他各时间点均有统计学意义(均为P<0.05);两组各个时间点相比较,差异均有统计学意义(均为P<0.05)。ADP酶组织化学染色显示:对照组视网膜血管自视盘发出的大血管向四周呈放射状分布,毛细血管分布均匀;糖尿病组大鼠16周视网膜出现视盘周围血管灌注降低,周边部部分毛细血管迂曲及血管闭塞。结论糖尿病大鼠视网膜中GLUT1表达增强,并随病程进展逐渐增高。出现明显视网膜微血管改变时,显著增高。 Objective To investigate the expression of glucose transporter-1 (GLUT1) in retina of rats with early-period diabetes, and explore the influence of GLUT1 on retina of early-period diabetes. Methods A total of 80 healthy male Wistar rats with 12 weeks old were used. Diabetic experiment model in 40 rats were induced by streptozotocin (4 cases died for server infection) ,which were injected into the left ab- domen, and the other 40 rats were injected tales doses citrate buffer solution as control group. The blood glucose and glycosylated hemoglobin of rats in two groups were exam- ined at 4 weeks,8 weeks,12 weeks and 15 weeks,9 cases in each time point of diabetic group and 10 cases of control group. The immunobistochemistry method was used to measure the expression of GLUT1 in the retina. ADP enzymohistochemistry stain meth- od of retinal stretched preparation was used to observe the change of retinal blood ves- sels. Results Blood glucose and glycosylated hemoglobin in diabetic group were sig- nificantly higher than those in control group. Immunohistochemical staining and light microscopic morphology showed that the cells in retinal ganglion cell layer arranged in great disorder at 16 weeks in diabetic group with no bleeding, retinal inner nuclear layer was edema,inner and outer nuclear layer cells arranged in disorder. The positive stal* ning particles of GLUT1 expression in the diabetic group was more than that in the con- trol group, there were statistical differences at each time points between two groups ( all P 〈 0.05 ). AOD values of GLUT1 expression in the diabetic group were 63. 903 3 ± 42. 245 3 at 4 weeks, 96.369 1 ± 18.697 2 at 8 weeks, 102.002 8 ± 23. 998 9 at 12 weeks and 130.408 7 ±72.745 1 at 16 weeks,respectively. The differences of GLUT1 AOD was not significant between 8 weeks and 12 weeks (P 〉 0.05 ) ,the others were all significant ( all P 〈 0.05 ). ADP enzymohistochemistry staining method showed that the large ves-sels from the optic disc were around scattered,
