摘要
目的:评价一种直接从标本中快速检测耐甲氧西林金黄色葡萄球菌(MRSA)的实时荧光PCR方法。方法:临床送检标本中筛选出46份金黄色葡萄球菌阳性的标本,进行实时荧光聚合酶链反应(PCR)检测,并与全自动细菌鉴定仪(VITEK-2Compact)/头孢西丁纸片扩散法进行比较,评价实时荧光PCR方法的敏感性及特异性。结果:VITEK-2Compact分离出金黄色葡萄球菌46株,其中MRSA 12株,与头孢西丁纸片扩散法结果一致。实时荧光PCR法检测出金黄色葡萄球菌46株,其中MRSA 13株,敏感性及特异性分别为100%(95%CI,75.75,100),97.06%(95%CI,85.08,99.48)。2种方法有很好的一致性[Kappa=0.945(95%CI,0.839,1.000)]。结论:实时荧光PCR法是一种能从标本中直接检测MRSA的快速,敏感的方法。
Objective:To evaluate a rapid method for the detection and identification of methicillin-resistant Staphylococcus aureus(MRSA)from clinical specimens.Method:46Staphylococcus aureus positive clinical samples were detected by real-time PCR.VITEK 2Compact and cefoxitin disk diffusion method were used as golden standard to evaluate the sensitivity and specificity of the real-time PCR method.Result:VITEK 2Compact and cefoxitin disk diffusion method identified 46 Staphylococcus aureus strains,of which 12 strains were MRSA.Realtime PCR detected 46 Staphylococcus aureus,including 13 MRSA.The sensitivity and specificity were 100%(95% CI,75.75,100)and 97.06%(95% CI,85.08,99.48),respectively.Real-time PCR method showed very good consistency with the reference method[Kappa=0.945(95% CI,0.839,1.000)].Conclusion:Real-time PCR method may be a reliable and fast way to detect and identify MRSA directly from the clinical specimen.
