摘要
目的观察常用清热凉血药(生地、紫草、丹皮)煎剂对银屑病小鼠阴道上皮有丝分裂及尾部鳞片表皮颗粒层的影响及其量效关系,并探讨其作用机制。方法采用灌胃雌激素刺激小鼠阴道上皮组织增生的方法制作银屑病小鼠动物模型,直接利用小鼠尾部鳞片模型,给予不同剂量(1.93、3.86、7.72 g/kg)的3种清热凉血药进行干预,14 d后计数小鼠阴道上皮有丝分裂指数和小鼠尾部颗粒层鳞片数。结果与对照组相比,清热凉血药生地、紫草、丹皮的各剂量组对银屑病样表皮过度增殖均有不同程度的抑制作用(P<0.05);均能促进表皮颗粒层形成(P<0.05),但3味药之间的作用效果无明显差异,同一味中药不同浓度之间亦无明显量效关系。结论清热凉血药生地、紫草、丹皮可能通过抑制上皮细胞有丝分裂和促进表皮颗粒层生成发挥治疗银屑病的作用。
Objective To investigate the effects of three heat-clearing and blood-cooling traditional Chinese herbal medicine( Shengdi,Zicao,Danpi) on mouse models of psoriasis,including estrogeninduced vaginal epidermis and the mouse-tails ' granular flakes,and to explore their mechanism of action. Methods The induced vaginal epidermis mouse models were established by feeding the mouse with estrogen to facilitate the formation of their vaginal epithelia. Whereas the mouse-tails' models were built by using mouse tail that were born without stratum granulosum. The researcher gave three different heat-clearing and blood-cooling traditional Chinese medicinals to mouse models for 2 weeks at different dosages( 1. 93,3. 86,7. 72 g / kg),and then counted the mitotic index of the mouse's vaginal epithelia and the number of mouse-tails' granular flakes. Results Compared with the control groups,each dose of three medicinals inhibited mouse vaginal epithelial mitosis( P〈0. 05); they also facilitated the formation of the granular layers in the flakes of the mouse-tails( P〈0. 05),but the there were no marked differences in the effects among these three medicinals and there was no direct dose-effect relation in each medicinal. Conclusion Three heat-clearing and blood-cooling traditional Chinese medicine( Shengdi,Zicao,Danpi) could help alleviate psoriasis through inhibiting mouse vaginal epithelial mitosis and facilitating the formation of the granular layers in the flakes of the mouse-tails.
