摘要
目的研究益肾解毒汤对单侧输尿管梗阻(UUO)大鼠肾组织氧化应激的干预作用。方法用UUO法建立慢性肾衰竭大鼠模型,SD雄性大鼠随机分为假手术组、模型组、益肾解毒汤组和科素亚组,于造模后7d、14d各组随机选取10只检测血肌酐(Scr),尿素氮(BUN),24h尿蛋白定量(24h Upro),肾组织匀浆超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽(GSH)。结果与假手术组比较,模型组肾组织SOD、GSH明显降低(P<0.01),Scr、BUN、24h Upro、MDA明显升高(P<0.01);益肾解毒汤组与模型组比较,肾组织SOD、GSH明显升高(P<0.01),Scr、BUN、24h Upro、MDA明显下降(P<0.05);益肾解毒汤组与科素亚组比较,肾组织SOD、GSH明显升高(P<0.05),Scr、BUN、24h Upro、MDA明显下降(P<0.05)。结论在肾间质纤维化的整个发展过程中,都有氧化应激反应的参与,益肾解毒汤能有效减轻慢性肾衰竭大鼠的氧化应激反应,进而可能起到延缓肾间质纤维化的作用。
Objective To explore effect of Yishenjiedu decoction( YSJD) on oxidation stress reaction in rat kidney after unilateral urethral obstruction. Methods Sprague Dawley male rats were randomly divided into 4 groups: the sham- operation group,the model group,the YSJD group,and the Lostartan group. Each group was randomly choosen 10 rats after establishing model for 7days and 14 days to detect. blood urea nitrogen( BUN) and creatinine( Scr). Collecting 24 h urine to detect urinary protein( 24 h Upro). Malondialdehyde( MDA),superoxide dismutase( SOD),glutathione( GSH) contents in obstructed kidney tissue of rats were tested. Results Compared with those in the sham-operation group,SOD and GSH contents in the model group were decreased( P 0. 01),while the content of 24 h Upro,BUN,Scr and MDA increased( P 0. 01). Compared with those in the model group,SOD and GSH contents in the YSJD group increased( P 0. 01),while the content of 24 h Upro,BUN,Scr and MDA decreased( P 0. 05). Compared with those in the Lostartan group,SOD and GSH contents in the YSJD group increased( P 0. 05),while the content of 24 h Upro,BUN,Scr and MDA decreased( P 0. 05). Conclusion Oxidation stress reaction exists in the onset and development of renal interstitial fibrosis. Yi shen jie du decoction may degrade renal interstitial fibrosis by suppressing oxidative stress reaction in rats after UUO.
