摘要
目的通过大鼠股骨骨髓原位灌注模型研究体内使用岩藻依聚糖上调骨髓内中性粒细胞趋化因子受体4[chemokine(C-X-C motif)receptor 4,CXCR4]表达的可能性,并探索其对于体外循环相关骨髓中性粒细胞快速释放的调节作用。方法 SD大鼠12只随机分为岩藻依聚糖灌注组(F组,n=6)和对照组(C组,n=6)并建立股骨骨髓原位灌注模型。F组大鼠模型灌入岩藻依聚糖溶液(灌入总量6 ml,灌注1 h),对照组灌入缓冲盐溶液。1 h后分别解剖冲洗收集两组大鼠骨髓细胞,用流式细胞仪比较两组冲洗液中性粒细胞CXCR4表达强度。再以SD大鼠18只随机分为3组,每组6只,并建立股骨骨髓原位灌注模型。分别分为岩藻依聚糖溶液灌注组,,(F组)、岩藻依聚糖溶液+AMD3100灌注组(F+AMD3100组)和对照组(C组)。每组骨髓均在灌入体外循环大鼠血浆之前分别灌入岩藻依聚糖溶液、岩藻依聚糖溶液+AMD3100和缓冲盐溶液。继以缓冲盐溶液灌注40 min后收集各组灌出液并计数其中中性粒细胞总数。结果 F组冲洗液中性粒细胞CXCR4(+)比例和表达强度分别为4.71%±0.21%和161.3±7.8,C组冲洗液中性粒细胞CXCR4(+)比例和表达强度分别为1.11%±0.11%和58.4±6.5,,,F组两项指标均明显高于C组,差异有统计学意义(P<0.05)。F组、F+AMD3100组和C组共收集到骨髓中性粒细胞数平均分别为(261 393.7±12 470.6)个、(872 635.2±10 430.6)个和(818 675.2±10 708.8)个,三组间两两比较差异均有统计学意义(P<0.05)。结论在SD大鼠股骨骨髓原位灌注模型中使用岩藻依聚糖可以有效上调骨髓中性粒细胞表面CXCR4的表达。使用岩藻依聚糖上调骨髓中性粒细胞表面CXCR4的表达可以有效减少体外循环血浆刺激下骨髓中性粒细胞的释放,且这一作用可被AMD3100取消。
Objective Based on the rat in situ perfusion system, to explore the effect of up-regulating Chemokine(C-X-C motif) receptor 4(CXCR4) expression on bone marrow neutrophils in modulating its ECC-related rapid release. Methods Twelve SD rats were randomly divided into fucoidan perfusion group(F, n=6) and control group(C, n=6) after in situ perfusion system establishment. Rats in F group received perfusion of fucoidan solution(total volume 6 ml, 1 h) and C group received buffer only. Femurs from two groups were dissected after one-hour perfusion and bone marrow tissues were collected. The neutrophil CXCR4 expression in two groups were compared using flowcytometry.,Eighteen SD rats were randomly divided into fucoidan perfusion group(F, n=6), fucoidan and AMD-3100 perfusion,group(F+AMD3100, n=6) and control group(C, n=6) after in situ perfusion system establishment. Rats received desired interventions before stimulation from ECC plasma. After that, 40-min perfusions of buffer were added and total counts of neutrophil in perfusates were compared. Results The percentages of CXCR4(+) cell and CXCR4 expression fluorescence in F group were 4.71%±0.21% and 161.3±7.8 respectively while the values were 1.11%±0.11% and 58.4±6.5respectively in C group. Values in F group were both significantly higher than those in C group(P0.05). The total counts,,of neutrophil in perfusates from F group, F+AMD3100 and C group were 261 393.7±12 470.6, 872 635.2±10 430.6 and818 675.2±10 708.8, respectively. Statistically differences were observed between each other(P0.05). Conclusion Bone marrow neutrophil CXCR4 expression of SD rat could be effectively up-regulated by perfusion of fucoidan within the in situ perfusion system. ECC-plasma-stimulated bone marrow neutrophil release in rat could be inhibited by fucoidan induced up-regulation of neutrophil CXCR4 expression, and this inhibition effect could be canceled by AMD-3100 intervention.
