摘要
目的观察五色梅总黄酮对Ⅱ型胶原诱导关节炎大鼠的治疗作用,并探讨其作用机制。方法采用Ⅱ型胶原诱导关节炎大鼠模型,设正常对照组(给予等体积纯化水)、模型对照组(给予等体积纯化水)、阳性对照组(给予布洛芬50 mg·kg-1)、五色梅总黄酮大剂量组(100 mg·kg-1)、五色梅总黄酮小剂量组(25 mg·kg-1);各组每日1次,给药21 d,观察大鼠足肿胀率及关节炎指数的变化,测定大鼠炎性组织液中的白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、超氧化物歧化酶(SOD)活性和丙二醛(MDA)的水平。结果五色梅总黄酮能降低Ⅱ型胶原诱导关节炎大鼠足肿胀率并降低关节炎指数,在造模后第17天,五色梅总黄酮大剂量组,足趾肿胀率由模型对照组(61.85±15.26)%降低到(47.04±14.20)%,关节炎指数由模型对照组(4.76±1.27)下降到(3.69±0.53)(P<0.05);并能显著降低炎性组织液中IL-1β、TNF-α的水平,五色梅总黄酮大剂量组,IL-1β水平由模型对照组(888.62±73.28)μg·m L-1下降到(805.75±98.51)μg·m L-1,TNF-α水平由模型对照组(928.80±81.37)μg·m L-1下降到(820.23±126.40)μg·m L-1(P<0.05);能显著降低炎性组织液中MDA水平,提高SOD的活性,五色梅总黄酮大剂量组,MDA水平由模型对照组(26.28±4.94)nmol·m L-1下降到(21.56±5.01)nmol·m L-1,SOD活性由模型组(159.68±51.18)U·m L-1提高到(212.08±54.65)U·m L-1(P<0.05)。结论五色梅总黄酮有效改善Ⅱ型胶原诱导关节炎大鼠关节炎症状,其机制可能与改善炎性组织液中IL-1β、TNF-α水平及抗氧化有关。
Objective To observe the antiarthritic effect and the mechanism of total flavonoids of Lantana Camara on rats with Collagen H-induced arthritis. Methods Arthritis rat model was established with Collagen ]]. The rats with arthritis were divided into five groups: normal control group, model control group, ibuprofen group (50 mg · kg-1 ), large dose ( 100 mg · kg-1 ) and small dose (25 mg · kg-1 ) of total flavonoids of Lantana Camara group. The medicines were given once daily for 21 days. The changes of paw swelling and arthritis index were observed. Levels of interleukinl[3 (IL-1 [3), tumor necrosis factor α (TNF-α), superoxide-dismutase (SOD) and malonic dialdehyde (MAD) in inflanmmtory tissue fluid were determined. Results Compared with model control group, the total flavonoids of Lantana Camara inhibited paw edema and decreased arthritis index in arthritis rats significantly. Rate of paw edema was (61.85±15.26)% and (47.04±14.20)% in model control group and large dose group, respectively. Arthritis index was 4.76±1.27 and 3.69±0. 53 in model control group and large dose group, respectively (P〈0. 05 ). As compared with model control group, total flavonoids of Lantana Camara in large dose significantly decreased levels of IL-I[3 [ (888. 62±73.28)μ·mL-1 vs. (805.75±98.51) μ·mL-1, P〈0.05] and TNF-α [(928.80±81.37)μ·mL-1 vs. (820.23±126.40)μ·mL-1, P〈0.05]; the level of MDA was significantly decreased and the activity of SOD significantly improved in large dose of total flavonoids of Lantana Camara group [ (26.28±4.94) nmol· mL-1 vs. (21.56-±5.01) nmol· mL-1, ( 159.68±51. 18) U · mL-1vs. (212.08±54.65) U· mL-1 , respectively, P〈0.05 ]. Conclusion The total flavonoids of Lantana Camara has therapeutic effect on arthritis rats. The anti-arthritic effect is closely related to serum levels of IL-1 β, TNF-α, and antioxidation ability.
