摘要
目的探究不同病理分级的宫颈鳞状细胞癌患者血清及癌组织中T细胞免疫球蛋白黏蛋白3(Tim-3)及其配体半乳凝素9(Gal-9)表达,分析其临床意义。方法收集17例上皮瘤样病变(CIN),38例含低(PSCC)、中(MSCC)、高分化(WSCC)宫颈鳞状细胞癌(SCC)组织以及血清样本,17例正常妇女血清,并依次分为CIN组、PSCC组、MSCC组、WSCC组以及正常组。分别采用蛋白质免疫杂交技术(Western blot)以及q RT-PCR技术检测CIN组、PSCC组、MSCC组以及WSCC组组织样本中Tim-3以及Gal-9的表达,采用酶联免疫吸附法(ELISA)检测CIN组、PSCC组、MSCC组、WSCC组以及正常组血清样本中Gal-9的表达水平。分别比较各组组织中Tim-3和Gal-9的表达以及血清中Gal-9的表达水平与宫颈鳞状细胞癌不同病理分级的相关性。结果 Western blot与q RT-PCR结果均显示Tim-3和Gal-9在SCC各组组织中表达高于CIN组(P<0.05),且与病理分级呈正相关(r=0.738、0.671,P<0.05)。ELISA结果亦显示Gal-9在SCC各组血清中高于正常组以及CIN组(P<0.05),切与病理分级呈正相关性(r=0.687,P<0.05)。结论 Tim-3及其配体Gal-9的表达与宫颈鳞状细胞癌病理分级成正相关,Tim-3以及Gal-9可作为临床诊断及预后的有义指标。
Objective Our study investigates the expression of T cell immunoglobulin mucin 3(Tim-3) and its ligand galectin-9(Gal-9) in the serum and cancer tissue in patients with squamous cell carcinoma of the cervix of different pathological gradings and analyzes its clinical significance. Methods We collected the samples of tissues and serum of 17 cases of cervical intraepithelial neoplasia(CIN), 38 cases of squamous cell carcinoma(SCC) of the cervix including poorly differentiated squamous cell carcino-ma(PSCC) of the cervix, moderately differentiated squamous cell carcinoma(MSCC) of the cervix, well differentiated squamous cell carcinoma(WSCC) of the cervix, and 17 cases of normal women and grouped them as the CIN group, PSCC group, MSCC group,WSCC group, normal group in turn. The expressions of Tim-3 and Gal-9 in the tissues of CIN, PSCC, MSCC, WSCC group detected by Western blot and q RT-PCR were compared between the groups and the expressions of Gal-9 in the serum of the above groups and the normal group detected by ELISA were also compared between the groups so as to find the correlation between their expressions and pathological grading of squamous cell carcinoma of the cervix. Results The result of Western blot and q RT-PCR showed that Tim-3 and Gal-9 expression in the tissues of SCC groups were higher than that in the CIN group(P〈0.05), and Tim-3and Gal-9 had positive correlation with pathological gradings(r=0.738, 0.671, P〈0.05). The result of ELISA showed that the expression of Gal-9 in the serum of SCC groups was higher than that in the normal group and CIN group(P〈0.05), and Gal-9 had positive correlation with pathological gradings(r=0.687, P〈0.05). Conclusion The expression of Tim-3 and its ligand Gal-9 has positive correlation with the pathological gradings of squamous cell carcinoma of the cervix. Tim-3 and Gal-9 can be the significant indicators of clinical diagnosis and prognosis.
