摘要
目的:研究西黄丸对人结直肠癌细胞SW480凋亡的影响及其作用机制的初步探讨。方法:以人结直肠癌细胞SW480为研究对象,MTT法检测不同剂量西黄丸含药血清对SW480细胞活性的影响,并用流式细胞术检测西黄丸含药血清引起的细胞凋亡率改变;用Western blot的方法检测Bax和Bcl-2蛋白表达水平,初步探讨其凋亡机制。结果:西黄丸含药血清可浓度依赖性降低SW480的细胞活力并增加细胞凋亡率,Western blot结果显示西黄丸含药血清增加了Bax蛋白表达,抑制Bcl-2蛋白表达。结论:西黄丸含药血清可抑制SW480细胞活力、诱导细胞凋亡并减小Bcl-2、Bax蛋白表达比例,其机制可能与以线粒体为核心的凋亡途径有关。
Objective: To investigate the effect of Xihuang Pills drug serum on the apoptosis of human colorectal cancer SW480 cells and the mechanism of Xihuang Pills. Methods: Taking the human colorectal cancer SW480 cells as the study targets, MTT assay and flow cytometry were used to test the effects of different doses of Xihuang Pills drug serum on SW480 cells viability and apoptosis changes of SW480 cells respectively. The protein expression of Bax and Bcl-2 were detected by Western blot assay, to explore the apoptosis mechanism. Results: Cells viability dose-dependently decreased in SW480 cells after the treatment with Xihuang Pills drug serum, while cells apoptosis rate increased. The Western blot results showed that the Xihuang Pills drug serum enhanced the protein expression of Bax, and suppressed the expression of Bcl-2. Conclusion: Xihuang Pills drug serum could reduce the SW480 cells viability, induce apoptosis and down-regulate the proportion of protein expression of Bcl-2, Bax in SW480 cells. The mechanism may be related to the mitochondrial apoptotic pathway.
