尼莫地平对脑缺血再灌注损伤大鼠细胞凋亡及细胞色素C表达的影响 被引量:1

Effect of Nimodipine on Cell Apoptosis and the Expression of Cytochrome C in Rat Brain Tissue after Cerebral Ischemia Reperfusion

摘要 目的:探讨尼莫地平对缺血再灌注大鼠脑组织细胞凋亡和细胞色素C蛋白(cytochrome C,Cytc)表达的影响。方法:将大鼠随机分为假手术组、模型组、尼莫地平组,每组27只。采用线栓法栓塞大脑中动脉3h制作大鼠脑缺血再灌注模型;分别于6h、12h、24h用TUNEL法检测大脑皮层细胞凋亡、Western blot检测大脑皮层Cytc蛋白的表达。结果:TUNEL法和Western blot结果显示:(1)尼莫地平组各时间点神经细胞凋亡较模型组明显减少(P<0.01);(2)模型组Cytc蛋白各时间点表达较假手术组有显著性差异(P<0.01、P<0.05);(3)尼莫地平组各时间点较模型组有显著性差异(P<0.01)。结论:尼莫地平对脑缺血再灌注损伤保护机制与抑制Cytc蛋白表达有关。 Objective:To investigate the effects of nimodipine on cell apoptosis and the expression of cytochrome c in the brain tissue after cerebral arterial thrombosis in rats.Methods:Rats were randomly divided into three groups:the sham-operative group,the model group and the nimodipine group(n = 27 in each group).The model of middle cerebral artery occlusion for 3 h was induced by suture method.The apoptosis of perifocal cortex was detected by TUNEL method and the expression of cytochrome c was analyzed by Western blot at the end of 6 h,12 h and 24 h.Results The results of TUNEL and Western blot showed:(1)Compared with the model group,the apoptosis decreased obviously in the nimodipine group (P 〈0.01);(2)Compared with the sham group,the expression of cytochrome c in the perifocal cortex of the model group was significantly different(P 〈0.01);(3)the expression of cytochrome C was reduced significantly in the nimodipine group at every time compared with the model group(P 〈0.01).Conclusion:Nimodipine can protect the brain cells from damage after cerebral arterial thrombosis injury by adjusting the expression of cytochrome C.
