miRNA-16在系统性红斑狼疮患者外周血中的表达及作用

摘要 目的探讨miRNA-16表达在系统性红斑狼疮(SLE)发病中的作用。方法选取符合1997年美国风湿病学会修定的SLE诊断标准的SLE患者16例(SLE组),体检正常的健康对照者12名(健康对照组)作为研究对象。取研究人群外周血分离外周血单个核细胞(PBMC),提取和纯化总RNA,实时定量PCR检测miRNA-16的表达,以小分子RNAU6作为内对照,计算标准化后的2-△ct。值表示miRNA的相对表达含量。并分析其与SLE临床特征的相关性。结果SLE组PBMC中miRNA-16的表达水平919.87±715.45高于健康对照组413.63±330.69,差异有统计学意义(t=-2.497,P〈0.05);SLE活动组miRNA-16的表达水平1298.79±803.79高于稳定组540.95±350.15,差异有统计学意义(t=-2.445,P〈0.05);miRNA-16的表达水平与抗核小体抗体呈正相关(r=0.669,P=0.005),与红细胞沉降率呈正相关(r=0.608,P=0.012),与SLE活动指数评分呈正相关(r=0.530,P=0.035)。结论SLE患者PBMC中miRNA-16异常高表达,与狼疮活动有关。 Objective To investigate the expression of miRNA-16 in peripheral blood monoucleacell(PBMC) from systemilupuerythematosu(SLE) patients. MethodSixteen SLE patientwho meethe diagnosticriteriof SLE revised in 1997 American rheumatology and 12 healthy individualwere selected aousubjects. Theiperipheral blood were sampled. Total RNAwere extracted and purified. The level of miRNA-16 wadetermined by quantitative reverse transcription PCR(qRT-PCR). U6 waused ahousekeeping control. The amounof targemiRNwanormalized relative to the amounof U6 ( AC= ACmiRNA-ACtU6). Relative expression levelwere expressed a2-△ct. ResultThe expression level of miRNA-16 in the SLE patientwa919. 87 ± 715.45, significantly highethan thain the healthy control group(413.6 3 ± 330. 69;= -2. 497, P 〈 0. 05 ). And miRNA-16 expression in SLE active group wa1 298.79 ± 803.79, significantly highethan thain SLE stable group( 540. 95 ± 350. 15 ; = - 2. 445, P 〈 0.05 ). The level of miRNA-16 warelated with Anu(r=0. 669,P=0. OO5),ESR(r=0. 608,P=0. 012) and SLEDAI(=0.530,P =0.035). Conclusion The expression of miRNA-16 ihigh in SLE patientand iirelated with SEE activity.
