摘要
基于表达谱数据分析,根据公共数据库中的序列信息设计引物,通过RT-PCR扩增及克隆,从耐冷的东乡野生稻(Dongxiang Oryza rufipogon Griff.)中获得了在冷胁迫条件下高表达的热激转录因子BGIOSGA004402-DX的全长c DNA,与冷敏感水稻品种93-11的对应位点序列进行氨基酸同源比较分析,结果表明:该基因在2个不同材料中长度一样,都为249个氨基酸,具有99%的序列一致性,但存在2个氨基酸的差异,一个是第127号氨基酸,另一个是第155号氨基酸。
The full length c DNA of high expression of heat shock transcription factor BGIOSGA004402-DX was obtained and cloned from cold-tolerant Dongxiang wild rice( Oryza rufipogon Griff) based on analysis of expression profile data by reverse transcription polymerase chain reaction( RT-PCR) with the primers designed on the basis of sequence information in public database. The comparison results of amino acid sequence with corresponding locus of cold-sensitive rice variety 93-11 indicated that the length of the gene had the same size of 249 amino acids in two different materials,with 99% of the sequence of consistency. However,the difference of two amino acids in the gene was detected in two different varieties. One was No. 127,and the other was No. 155.
