摘要
目的建立大鼠糖尿病肾病模型,探讨白藜芦醇其肾功能保护的机制。方法建立大鼠糖尿病肾病模型,生化分析法检测血糖,酶联免疫法测定24小时尿白蛋白排泄率,肌酐清除率,Western blot法检测自噬标志蛋白LC3-II和Beclin1水平变化。结果白藜芦醇治疗对糖尿病肾病大鼠血糖无明显影响(P>0.05),但可明显降低糖尿病肾病大鼠肌酐清除率和24h尿白蛋白量(P<0.05),提高LC3-II和Beclin1蛋白表达。结论白藜芦醇可通过活化自噬功能发挥对糖尿病大鼠肾脏功能的保护作用。
Objective To investigate the mechanism of renal protective effects of resveratrol on diabetic rats.Meth-ods Establishing DN rat model by streptozotocin biochemical analysis of blood glucose,measured 24-hour urinary al-bumin excretion rate by enzyme-linked immunosorbent assay,creatinine clearance rate,RT-PCR was used to detect mi-tochondrial fission,fusion gene transcripts levels,mitochondrial membrane potential detection.Results Resveratrol treatment had no significant effects on blood glucose in DN rats (P〉0.05),but significantly reduced creatinine clear-ance and 24h urinary albumin(P〈0.05),and increased Beclin1 and LC3-Ⅱ protein expression.Conclusion Resveratrol plays a protective role on renal function in DN rats by affecting autophagy function.
