藏族药翼首草正丁醇部位提取物体外抑制人肝癌Hep3B细胞增殖和侵袭转移 被引量:10

Inhibits Human Liver Hep3B Cell Proliferation and Invasion and Metastasis of n-butanol Part of Pterocephalus hookeri in Vitro

摘要 目的:本研究旨在体外筛选出藏族药翼首草正丁醇部位提取物(YSC-ZDC)抑制人肝癌Hep3B细胞侵袭转移的有效萃取部位,并对其抗肿瘤作用机制进行初步探讨。方法:取对数生长期Hep3B细胞,调整细胞密度为4×10^4/m L(MTT法)或2.5×105/m L(Western blot,heochst 33258实验),1×10^5/m L(划痕实验),5×10^4/m L(Transwell实验),培养24,48,72 h后分别加入不同质量浓度的药物(50,100,200 mg·L^-1),分组设3-4个复孔。通过MTT法检测YSC-ZDC对于Hep3B细胞增殖的影响,形态观察该药对Hep3B细胞凋亡的影响。划痕实验和Transwell小室检测YSC-ZDC对Hep3B细胞侵袭、迁移的影响;Western blot检测侵袭转移相关信号通路分子表达的变化。结果:YSC-ZDC 50,100,200 mg·L^-1作用于Hep3B细胞24,48,72 h后可明显抑制其增长。50,100,200 mg·L^-1YSC-ZDC作用于Hep3B细胞24 h后可明显抑制其侵袭和迁移;YSC-ZDC可明显影响上皮-间质转化(EMT)相关蛋白E-钙黏素,N-钙黏素(E-cadherin,N-cadherin)和纤维连接蛋白(FN)的表达(P〈0.05)。结论:YSC-ZDC能显著抑制Hep3B细胞的增殖和侵袭转移,并诱导Hep3B细胞凋亡,其作用机制可能与EMT有关。 Objective: This study sought to investigate whether n-butanol part of Pterocephalus hookeri (YSC-ZDC) in vitro inhibits the invasion and metastasis of human liver cancer Hep3B cells effective extraction, and its antitumor mechanisms were discussed. Method: Hep3B cell proliferation was assayed by MTT method, the fluorescent staining was used to observe the effect of YSC-ZDC on Hep3B cell apoptosis. Scratch test and Transwell were used to detect invasion and migration of Hep3B cell by YSC-ZDC, Western blot was uswed to detect related signaling pathways. Result: YSC-ZDC 50, 100, 200 mg ·L^-1 24, 48, 72 h after treatment could obviously inhibit proliferation. 50, 100, 200 mg ·L^-1 YSC-ZDC after 24 h could obviously inhibit the invasion and migration; YSC-ZDC could significantly affect the epitheliai-mesenchymel transitions (EMT) related proteins. Conclusion: YSC-ZDC can significantly inhibit Hep3B ceils proliferation and invasion and metastasis, and induce the Hep3B cell apoptosis. Its mechanism may be related to EMT.
