摘要
目的:观察丁苯酞(NBP)预处理对急性低压低氧小鼠空间记忆功能、脑组织一氧化氮(NO)和内皮素-1(ET-1)含量的影响。方法:42只成年昆明小鼠被随机分为3组,即常压常氧对照组、低压低氧对照组和低压低氧实验组,通过Morris水迷宫实验的逃跑潜伏时间和平均速度评估小鼠空间记忆功能,并检测海马组织中的NO和ET-1含量。结果:与常压常氧组比较,低压低氧对照组和低压低氧实验组的潜伏期明显延长,平均速度显著变慢,低压低氧对照组脑组织NO和ET-1含量均显著增加,低压低氧实验组脑组织ET-1含量显著增加(P<0.05或<0.01);与低压低氧对照组比较,低压低氧实验组的潜伏期显著缩短,平均速度显著增快,脑组织ET-1含量显著减少(P<0.05)。结论:①低压低氧可致小鼠空间记忆功能受损,这可能与低压低氧所致的小鼠脑组织NO和ET-1异常有关;②丁苯酞预处理可以减轻低压低氧对小鼠记忆功能的损害,可能与丁苯酞减轻低压低氧所致的小鼠脑组织ET-1异常有关。
Objective: To observe the effects,; of butylphthalide (dl--3 --n- butylphthalide, NBP) preconditioning on spatial memory and the levels of nitric oxide (NO) and endothelin (ET--1) of the brain tissue in mice exposed to acute hypobaric--hypoxia. Methods: Forty--two Kunming mice were divided randomly into 3 groups, normobaric normoxia control group, hypobaric hypoxia control group and hypo- baric hypoxia experimental group. Escape latence and average velocity of Morris water maze test were em- ployed to evaluate spatial memory and the content of NO and ET--1 in the brain. Results: Compared to normobaric normoxic control group, a significant increase in escape latence and decrease in average velocity were found in hypobaric hypoxia control and experimental group (P〈0.05 or P〈0.01). Compared to normobaric normoxic control group, a significant increase in content of ET--1 and NO in the brain were found in hypobaric hypoxia control group and a significant increase in content of ET-- 1 were found in hy- pobaric hypoxia experimental group( P〈0.05 or P〈0.01). Compared to hypobaric hypoxia control group, a significant decrease in escape latence and a significant increase in average velocity and content of ET--1 in the brain were found in hypobaric hypoxia experimental group (P〈0. 05). Conclusion: The spatial memo- ry could be impaired by exposing to acute hypobaric--hypoxia, which may be related to the elevation of the levels of NO and ET-- 1 in brain tissue. Butylphthalide preconditioning can partially reverse of memory im- pairment caused by exposing to acute hypobaric--hypoxia, which may be related to the depression of the levels of ET--1 in brain tissue.
