EV71感染手足口病患儿外周血中Treg细胞、细胞因子和NSE的变化及其意义 被引量:3

摘要 目的探讨EV71感染手足口病患儿外周血中Treg细胞、细胞因子和NSE的变化,分析其在EV71感染手足口病患儿发病机制中的作用及意义。方法共采集EV71感染手足口病患儿97例(轻症组65例,重症组32例)和健康儿童50例(对照组)外周血,分别用流式细胞仪检测CD4+CD25+Foxp3+Treg,ELISA法测定IL-6、IL-10、TNF-α的含量,化学发光免疫分析仪检测血清中NSE水平,采用SPSS软件对所得数值进行统计学分析。结果与轻症组和对照组比较,重症组患儿外周血中CD4+CD25+Foxp3+Treg水平显著降低(P<0.05),IL-6、TNF-α、NSE水平显著增高(P<0.05);重症组患儿外周血中IL-10水平高于对照组,差异具有统计学意义(P<0.05);与对照组比较,轻症组患儿外周血中TNF-α、NSE表达水平显著升高(P<0.05)。结论观察EV71感染手足口病患儿外周血中CD4+CD25+Foxp3+Treg细胞、IL-6、IL-10、TNF-α、NSE水平的动态变化,可以为EV71型重症手足口病患儿早期鉴别诊断和治疗提供理论依据。 Objective To explore the change of Treg,cytokines( IL-6,IL-10,TNF-α) and neuron specific enolase( NSE)in the peripheral blood of patients with Enterovirus 71( EV71)-related hand-foot-mouth diseases( HFMD),and to analyze their role in the pathogenesis of EV71-related HFMD. Methods The peripheral blood of 97 cases of EV71-related HFMD( mild group,n = 65; serious group,n = 32) and 50 healthy children( control) were prepared to measure CD4^+CD25^+Foxp3^+Treg by flow cytometry( FCM),levels of IL-6,IL-10 and TNF-αby enzyme-linked immunosorbent assay( ELISA),and level of NSE in serum by Chemiluminescence immunoassay analyzer. SPSS software was employed for the statistical analysis. Results As compared with the control group and the mild group,the level of CD4^+CD25^+Foxp3^+Treg in the serious group reduced( P〈0. 05),the levels of IL-6,TNF-α and NSE in the serious group increased( P〈0. 05); the level of IL-10 in the serious group was higher than that in the control group,the difference was significant( P〈0. 05); the levels of TNF-α and NSE in the mild group increased obviously as compared with the control group( P〈0. 05). Conclusion The dynamic monitoring of CD4^+CD25^+Foxp3^+Treg,IL-6,IL-10,TNF-α and NSE in peripheral blood of EV71-related HFMD patients should provide significant theoretical basis for the prevention and treatment of severe EV71-related HFMD patients.
