摘要
目的:在HPLC法测定人血浆中氯氮平浓度的基础上建立UFLC法同时测定氯氮平及奎硫平浓度。方法血浆样品经适当处理,色谱柱为shim-pack XR-ODSш柱,流动相为0.4%三乙胺溶液-甲醇(45∶55,v/v),流速0.4 mL/min,检测波长257 nm。结果测定方法在50-1600μg/L范围内,氯氮平、奎硫平线性关系良好(氯氮平r=0.9999,喹硫平r=0.9993);日内、日间相对标准误差(RSD)在3.0%-6.5%。结论本方法快速、简便、准确,可用于血药浓度监测。
Objective In the HPLC method for the determination of human plasma clozapine concentration on the basis of the establishment of UFLC method for simultaneous determination of clozapine and quetiapine. Methods Plasma samples with appropriate treatment, the column was shim-pack XR-ODSшcolumn, the mobile phase was 0.4%three triethylamine solution-methanol(45∶55, V/V), the lfow rate was 0.4 mL/min, the detection wavelength was 257 nm. Results Method of determination in 50-1600μg/L range, clozapine, quetiapine good linear correlation(clozapine, r=0.9999, quetiapine, r=0.9993);days, day relative standard deviation (RSD)in 3.6%-5.7%between. Conclusion The method is quick, simple, accurate, and can be used for monitoring of serum drug concentration.
