摘要
本研究从96对SRAP引物中筛选出22对多态性高、稳定性好的引物,用于山楂资源的扩增,获得多态性位点,并采用NTSYS软件进行山楂资源的多样性分析。结果显示,每对引物组合产生8~20条谱带,其中多态性条带246条,占总带数的84.83%;57份种质的相似系数在0.66~0.92之间。聚类分析显示,在相似系数为0.748处57份山楂供试品种被划分为4个类群,并且来源地相同的品种间亲缘关系较密切。结果显示SRAP标记具较丰富的多态性,是进行山楂遗传多样性分析的有效分子标记。研究结果为分析山楂种质资源间亲缘关系及资源收集保存、开发利用等领域提供实践指导和理论依据。
In this study, 22 pairs of highly polymorphic and stable primers were selected from 96 pairs of SRAP primers for amplifying hawthorn resources to obtain polymorphic sites. And we used the NTSYS software to analyze the genetic diversity. Each pair of primers generated 8-20 bands, including 246 polymorphism bands that account for 84.83%. The similarity coefficient of the 57 accessions were from 0.66 to 0.92. The UPGMA cluster analysis showed that the 57 accessions were divided into 4 groups at 0.748 and the varieties came from the same source have the nearer genetic relationship. The result showed that the SRAP can generate abundant polymorphism and it is an effective molecular makers to analyze the hawthorn genetic diversity. The result might provide the theoretical basis and practice guidance for the genetic relationship between different hawthorn resources, the germplasm collection and development.
