摘要
【目的】研究番茄苗期响应盐胁迫生理指标及其变化规律。【方法】以盐敏感番茄品种M82和耐盐渐渗系IL7-5-5幼苗为材料,在室内利用1/2 Hoagland营养液进行水培养,通过200 mmol/L Na Cl胁迫处理,分别在加盐后0、1、12、24 h取样测定每个材料在各时间点叶片中丙二醛、脯氨酸和超氧化物歧化酶的含量,每个处理设3次重复。【结果】在盐胁迫条件下,耐盐番茄IL7-5-5较敏感番茄M82可保持较低的膜脂过氧化程度、较好的膜完整性以及较高的超氧化物歧化酶活性,从而保持细胞膜的有序性和稳定性;同时,耐盐番茄IL7-5-5可通过主动积累游离脯氨酸进行渗透调节,降低渗透势,提高渗透调节能力,从而保持相对较好的叶片水分状况。【结论】丙二醛、脯氨酸和超氧化物歧化酶的含量与活性可以作为番茄耐盐的评价指标,也可以通过检测这3种生理指标的变化筛选番茄耐盐品种。
【Objective】The project aims to study the physiological indexs and changing rules of salt-stress response at seedling stage in tomato.【Method】The seedlings of salt- sensitive variety M82 and the salt- tolerance introgression line IL7- 5- 5 were chosen as testing materials,and the seedlings were cultured indoors with hydroponics in 1 /2 Hoagland nutrient solution.. The contents of MDA,proline and SOD were respectively measured in leaves at 0,1,12,24 h after treatment with the 200 mmol / L Na Cl. Each treatment was repeated three times. 【Result】It was shown that under the salt stress,the salt- tolerance introgression line IL7- 5- 5 could maintain lower degree of membrane lipid peroxidation,better membrane completeness and higher SOD activity compared with the salt- sensitive variety M82,so that it could keep the membrane orderliness and stability; in the meantime,the salt- tolerance introgression line IL7- 5- 5 could increase its osmotic potential by active accumulating free proline to improve its capacity in osmosis regulation and its leaf water status. 【Conclusion】The content and activity of MDA,Pro and SOD could be used as evaluation index for salt- tolerance of tomato,and also used to screen the salt- tolerance variety of tomatoes.
