不同操作温度对小鼠卵母细胞体外受精的影响

摘要 目的:观察不同操作温度对小鼠体外受精以及后续胚胎发育的影响,探讨较合适的实验室受精及操作温度,为提高人类体外受精-胚胎移植(IVF-ET)技术提供实验依据。方法对IVF实验室温度共设置19℃、21℃、23℃、25℃、27℃、29℃及31℃温度点,选择10~12周龄(20~25g)的ICR雌性小鼠及雄性小鼠168只。每个温度点设自然受精组和体外受精组,随机分配雌鼠8只、雄鼠4只。在上述实验室温度下对小鼠自然受精卵团与体外受精卵团进行操作、观察,并统计受精率、卵裂率、囊胚形成率及囊胚孵出率。结果体外受精组中25℃组与19℃、21℃、29℃、31℃四组间比较受精率(94.83%、52.69%、78.98%、82.76%、66.67%)、囊胚形成率(87.12%、54.17%、68.75%、70%、56.56%)、囊胚孵出率(86.62%、46.15%、66.23%、67.857%、51.78%)有统计学差异(P<0.05),尤以19℃、31℃差异明显;体外受精组中25℃组与19℃、21℃、31℃三组间比较卵裂率(98.79%、81.82%、90.32%、89.83%)有统计学差异(P<0.05),尤以19℃、31℃差异明显;顺次两组间比较,19℃与21℃、29℃与31℃组受精率、囊胚形成率、囊胚孵出率有统计学差异(P<0.05);组内比较19℃、21℃、29℃、31℃组各自的受精率、卵裂率、囊胚形成率、囊胚孵出率有统计学差异(P<0.05),尤以19℃、31℃组差异明显。结论实验室温度在23℃~27℃之间时,小鼠卵母细胞体外受精组与自然受精组在受精率、卵裂率、囊胚形成率和囊胚孵出率等方面较其他温度具有显著优势,提示23℃~27℃为较合适的实验室受精及胚胎操作温度范围。 ObjectiveTo explore the suitable temperature by investigating the influence of different operating temperature on in vitro fertilization and subsequent embryonic development, in order to provide the potent experiment evidence for the IVF-ET technique.MethodsSeven temperature points(19℃,21℃,23℃,25℃,27℃,29℃,31℃)of the in vitro fertilization laboratory were set in our study. 10 to 12 weeks old ICR mouse of either gender weighting 20-25g were used as the object. The mouse (male=8,female=4)were randomly divided into nature fertilization group and in vitro fertilization group on each temperature point. The procedure was taken under the above seven different laboratory temperature points. The fertility rate,cleavage rate,blastocyst formation rate and blastocyst hatched rate were observed and counted.ResultsIn the in vitro fertilization group,the 25℃ group had statistically significant in comparison with the four groups of 19℃,21℃,29℃ and 31℃ in the fertilization rate(94.83%, 52.69%,78.98%,82.76%,78.98%),blastocyst formation rate(87.12%,54.17%,68.75%,70%,56.56%)and blastocyst hatched rate(86.62%,46.15%,66.23%,67.86%,51.78%),especially in the groups of 19℃ and 31℃,the difference were obvious. In the in vitro fertilization group,the cleavage rate had statistically difference(P〈 0.05)in the 25℃ group in comparing with the other three groups of 19℃,21℃ and 3℃(98.79%,81.82%,90.32%,81.82%). The difference was obvious especially in 19℃ and 31℃ groups. Progressive comparison between the two groups,the 19℃ group and 21℃ group,29℃ group and 31℃group had statistically difference(P〈 0.05)in the fertilization rate,blastocyst formation rate and blastocyst hatched rate. Comparison in the group,the 19℃,21℃,29℃ and 31℃ groups had statistically difference(P〈 0.05)in the fertilization rate,cleavage rate,blastocyst formation rate and blastocyst hatched rate,respectively,especially in 19℃ and 31℃ group,the difference were significantly.Concl
