摘要
研究了以木糖工业化生产的副产物木糖母液为碳源,利用地衣芽孢杆菌工程菌(Bacillus licheniformis)TCCC11843发酵生产碱性蛋白酶的工艺.对摇瓶发酵工艺进行优化,确定了接种种龄为12,h,接种量为8%,培养基初始p H为6.5,发酵温度为34,℃,在24,h一次补加4.0,m L质量分数为50%木糖母液,此时的发酵液酶活力峰值为7,838,U/m L;在此基础上,又在7,L发酵罐上实现了木糖母液的反馈补料,酶活力在80,h时达峰值,为14,912,U/m L.该结果为工业化生产碱性蛋白酶的发酵工艺优化提供了新的思路.
In this study,engineered Bacillus licheniformis TCCC11843,was used for the fermentation production of alkaline protease from a carbon source:xylose mother liquor,a by-product of xylose production. The optimized fermentation pa-rameters of alkaline protease production were:inoculum culture age was 12,h,initial pH 6.5,inoculum volume 8%, fermentation temperature 34,℃,and feeding 4.0,mL 50%xylose mother liquor at the 24th,h. Under such optimal fermenta-tion conditions,the maximum enzyme activity can reach 7,838,U/mL. Moreover,feedback feeding experiments using the 7,L fermenter resulted in the maximum enzyme activity of 14,912,U/mL at 80,h. These results are valuable for the optimal com-mercial production of alkaline protease.
