摘要
目的比较不同脾虚证慢传输型便秘(STC)小鼠模型的建立方法 ,优化其模型及评价方法。方法分别采用番泻叶+限水+控制饮食致STC小鼠模型(下称模型Ⅰ组)、游泳结合饥饱失常所致STC小鼠模型(下称模型Ⅱ组)及盐酸洛哌丁胺混悬液所致STC小鼠模型(下称模型Ⅲ组),比较各组小鼠一般行为学特征、首粒黑便传出时间、6 h排便数量及质量、肠道推进率、血清D-木糖含量及结肠黏膜组织病理学特征。结果与正常组相比,3种模型小鼠的粪便干结、质硬、数少、颗粒细小,首粒黑便传出时间显著延长(P<0.01),模型Ⅰ、Ⅲ组6 h排便数量和质量减少(P<0.01),模型Ⅰ、Ⅱ组血清D-木糖含量显著降低(P<0.01);但3种模型小鼠肠道推进率变化无统计学差异(P>0.05),其结肠黏膜正常或呈轻度炎症改变。结论模型Ⅰ组的一般行为学、首粒黑便传出时间、6 h排便数量和质量、血清D-木糖含量、结肠黏膜组织病理学检测可以作为评价脾虚证STC小鼠模型成功的客观指标。
Objective The different methods of slow transit constipation mouse models with spleen deficiency syndrome were compared to establish the optimization models and the evaluation system. Methods The rats were independently induced by senna + limited water + control diet(Model I), swimming + irregular diet(Model Ⅱ) and loperamide hydrochloride suspension(ModelⅢ). The behavioral characteristics, the excreted time of the first melena, 6 h defecation quantity and weight, small intestinal propulsion rates, serum D-xylose content, and pathological features of the colonic mucosa were compared in each group. Results Compared with the normal group, the stool of three model groups is dry, hard, less numbers, and small, the excreted time of the first melena was delayed(P〈0.01). The 6 h defecation quantity and weight in Model I andⅢ groups decreased(P〈0.01), the serum D-xylose content in Model I and Ⅱ groups reduced significantly(P〈0.01); But the small intestinal propulsion rates in the all model groups have no statistical differences(P〈0.05). The colonic mucosa of the all models was normal or showed mild chronic inflammatory changes. Conclusion The behavioral characteristics, the excreted time of the first melena, 6 h defecation quantity and weight, serum D-xylose content,and pathological features of the colonic mucosa in model groups could be considered as objective indicators to evaluate the success of STC mouse models with spleen deficiency syndrome.
