摘要 通过电子克隆与RT-PCR相结合的方法,获得一个新的烟草PHOT2基因。生物信息学分析表明,该基因c DNA全长3326bp,包含一个2889 bp的开放阅读框,共编码962个氨基酸。主要由无规则卷曲和α-螺旋构成蛋白的二级结构。具有典型的蓝光受体向光素功能结构域,即N端的两个PAS亚家族序列组成的光感受区和C端的Ser/Thr蛋白激酶区。此外预测到该蛋白存在几个潜在的磷酸化位点。 A new PHOT2 gene from tobacco( Nicotiana tabacum) was cloned by in silico cloning and RT-PCR technique. Bioinformatics analysis showed that Nt PHOT2 c DNA full length was 3326 bp,contained an open reading frame( ORF) of 2889 bp which it encoded 962 amino acids. The main motifs of the secondary structure are alpha helix and random coil. Nt PHOT2 protein had typical feature of blue-light receptor phototropin function domain,and contained two N-terminal light sensing domains belonging to PAS subfamily and C-terminal Ser / Thr kinase domain. Furthermore,several potential phosphorylation sites were predicted.
