血必净对脓毒症SD大鼠巨噬细胞移动抑制因子的影响 被引量:1

摘要 目的探讨血必净注射液对脓毒症大鼠巨噬细胞移动抑制因子(MIF)表达的影响及其对急性肾损伤的保护作用。方法参照Al-Abed Y等报道的方法建立盲肠结扎穿孔术诱导的小鼠脓毒症模型,按照随机数字法将60只SD大鼠分为对照组和血必净组;应用逆转录聚合酶链反应(RT-PCR)法和Westernblot法检测各时间点大鼠肾组织中MIF基因和蛋白的表达。同时采用生化法检测血清肌酐,酶联免疫吸附测定法(ELISA)检测血清MIF水平。数据结果采用SPSS16.0统计软件进行统计学分析。结果与对照组比较,血必净组大鼠肾组织MIF基因与血清MIF水平24 h、48 h明显下降(P<0.01),血清肌酐水平亦显著下降(P<0.01)。结论血必净可以通过下调MIF的合成与释放,有效地预防和保护肾组织炎症反应的发生、发展。 Objective To explore the Xuebijing influence on serum MIF expression of septic rats and prevention and cure function on acute kidney injury. Methods Sepsis model was duplicated by cecum ligation perforation( CLP). A total of sixty male Spargue- Dawley rats were divided by random number table method into two groups: the control group and the Xuebijing group. MIF mRNA and protein expressions in renal tissues of septic mice were assessed by reverse transcription- polymerase chain reaction( RT- PCR) and Western blotting. Biochemical method was used to detect serum creatinine levels. Results,through using SPSS16. 0 statistical software,did statistical analysis. Results Compared with control group,both the content of MIF in serum and the expressions of MIFmRNA in renal tissues of Xuebijing group came to descend obviously at 24 and 48 hours( P〈 0. 01); Scr level of Xuebijing group came to descend obviously( P〈 0. 01). Conclusion Xuebijing reduces the level of the kidney MIF gene and protein expression level,and it has an effectation on preventing and protecting acute kidney injury.
