3种不同睡眠剥夺小鼠模型的建立及其在中医药研究中的应用 被引量:9

摘要 目的建立3种小鼠睡眠剥夺模型并进行评价,探讨其在中医药研究中的应用。方法分别采用换笼、轻触、合并法(轻触+换笼)制作3种部分睡眠剥夺模型,观察睡眠剥夺对小鼠体征、自主活动及学习记忆的影响。结果睡眠剥夺后,与空白组比较,换笼、轻触、合并组小鼠平均体重差异明显,且合并组小鼠下降更为明显;在自主活动测试中,轻触组与合并组小鼠的自主活动次数、活动路程均明显减少(P<0.05);在水迷宫测试中,换笼、轻触、合并组小鼠上平台潜伏期时间延长,目标象限时间缩短(P<0.05,P<0.01),且合并组更为明显。结论合并法(轻触+换笼)睡眠剥夺效果明显优于换笼法和轻触法,且制作简单,还可同时对多只小鼠进行睡眠剥夺,经济适用可行可靠。同时可尝试在本模型的基础上叠加中医证候因素用于探讨中医药治疗失眠的病因病机。 Objective To establish three mouse models of sleep deprivation and evaluate, to explore its application in pharmaceutical re- search. Methods With cage changing, gentle handling and combined (both cage changing and gentle handling)procedures to establish par- tial sleep deprivation models, to observe the effects of sleep deprivation on spontaneous activity of mice, and the learning and memory of signs. Results After sleep deprivation, compared with the blank group, the average weight of the mice in cage changing, gentle handling and combined group significantly different, and the combined group were decreased more significantly; in the autonomic activity test, the number of autonomic activities and the movable distance of the mice in gentle handling and combined group were significantly reduced ( P 〈 O. 05 ), in the morris water maze test, mice in cage changing, gentle handling and combined group mounted the platform took longer time , shorter the time in the target quadrant ( P 〈 O. 05,P 〈 O. 01 ), the data of combined group' s mouse were more significantly different. Conclusion The combined (both cage changing and gentle handling)sleep deprivation method is obviously better than the sleep deprivation in cage changing and in gentle handling, and the manufacture is simple, which can be performed on many mice at the same time, and it is economic applicable and reliable. Meanwhile, we can try to add some TCM syndrome factors on this combined model to investigate the climate factors of Chinese medicine treatment of insomnia pathogenesis.
