摘要
目的:通过观察大黄对重症急性胰腺炎(SAP)大鼠肠黏膜Toll样受体(TLRs)的影响,探讨大黄对SAP肠黏膜屏障的保护机制。方法:将健康雄性SD大鼠随机分成假手术组、模型组、大黄治疗组。测定3组血清淀粉酶水平;胰腺、末端回肠组织病理评分;RT-PCR法检测回肠组织TLR2、TLR4 m RNA表达水平。结果:模型组肠黏膜TLR2、TLR4 m RNA的表达高于假手术组,大黄治疗组肠黏膜TLR2、TLR4 m RNA的表达低于模型组(P<0.05)。结论:大黄可降低重症急性胰腺炎TLR2、TLR4 m RNA的表达并改善肠黏膜屏障功能,从而抑制炎症反应的发展并减轻胰腺损伤。
Objective: To discuss the protection mechanism of rhubarb in intestinal mucosal barrier of rats with severe acute pancreatitis through observing the effects of rhubarb on the toll-like receptors in intestinal mucosal barrier. Methods: Healthy male SD rats were divided into sham operation group, model group and treatment group. The blood samples were collected for measuring the serum amylase. Histological changes of pancreas and terminal ileum were evaluated by the corresponding score system. Changes of expression of TLR2 and TLR4 mRNA were detected by RT-PCR in intestinal tissues. Results: The expression of TLR2 and TLR4 mRNA in rats of the model group was higher than that of the sham operation group, and the expression of TLR2 and TLR4 mRNA in rats of the treatment group was lower than that of model group (P〈0.05). Conclusion: Rhubarb could protect the intestinal mucosal barrier, inhibit the inflammatory response and reduce the pancreas injury by reducing the expression of TLR2 and TLR4 mRNA.
