摘要
目的:采用蛋白质组学技术探求脾阳虚证和脾阴虚证大鼠回肠组织蛋白的差异性表达。方法:36只大鼠随机分为正常对照组、脾阳虚证模型组和脾阴虚证模型组。采用劳倦过度、饮食不节与药物损伤相结合的方法建立脾阳虚证和脾阴虚证动物模型,通过模糊数学方法评价后分离回肠组织,后采用双向电泳法分离差异表达的蛋白,凝胶银染后经选点、酶切和质谱分析,用MASCOT软件在蛋白质数据库中搜索肽质量指纹谱数据并确定蛋白质。结果:脾阳虚证模型组回肠组织蛋白质图谱中有12个蛋白点表达异常(P<0.05),脾阴虚证模型组回肠组织蛋白质图谱中有7个蛋白点表达异常(P<0.05)。结论:脾阳虚证模型组与脾阴虚证模型组存在差异蛋白质组表达,脾阳虚证或与细胞骨架损伤和糖代谢异常的关系更为密切,脾阴虚证或与过氧化损伤的关系更为密切。
Objective: To identify the differential expression of ileum tissue proteins in rats with spleen-yang deficiency syndrome and spleen-yin deficiency syndrome by using proteomic method. Methods: Tirty-six rats were randomly divided into normal control group, spleen-yang deficiency syndrome model group and spleen-yin deficiency syndrome model group. The model rats were established by using exerting excessive, irregular diet and combined with drug injury. The ileum tissues were separated after evaluation by fuzzy mathematics method. The differential expression of proteins were separated by two dimensional electrophoresis method. After the gel was elected and digested by silver staining, and analyzed by Mass spectrometry, the peptide mass fingerprinting data were searched in protein database and identified the proteins by MASCOT software. Results: The differential expression of 12 proteins were found in spleen-yang deficiency syndrome model group while 7 proteins were found in spleen-yin deficiency syndrome model group (P〈0.05). Conclusion: The differentiai expression between the spleen-yang deficiency syndrome model group and the spleen-yin deficiency syndrome model group are existed. The spleen-yang deficiency syndrome is more closely related with damage of cytoskeleton and abnormal glucose metabolism, and spleen-yin deficiency syndrome is closely related with peroxidation injury.
