脾虚大鼠壁细胞胃泌素受体研究及黄芪作用观察 被引量:15

摘要 目的 :观察脾虚大鼠胃壁细胞胃泌素受体结合位点数的变化以及中药黄芪注射液对其的调控作用。方法 :脾虚模型采用利血平方法 ,治疗组给予黄芪注射液 ;大鼠胃黏膜壁细胞分离与纯化采用Lewin胃袋法及Percoll密度梯度纯化 ;壁细胞胃泌素受体结合位点数 (sites cell)的检测采用放射配基结合方法。结果 :利血平脾虚大鼠壁细胞胃泌素受体结合位点数为 2 71 2± 91 8,明显低于正常组 (6 19 1± 30 4 5 ,P <0 0 5 )。中药黄芪注射液对其有显著上调作用 (5 45 2± 2 0 5 3,P<0 0 1)。结论 :利血平脾虚大鼠胃壁细胞胃泌素受体结合位点数下降 ,中药黄芪注射液对其有明显上调作用 ,胃泌素受体与脾虚证关系密切。 AIM:To observe the gastrin receptor binding sites and the effects of Huangqi on which in spleen deficiency rats.METHODS:The spleen deficiency models were made with reserpine.The rats were treated by Huangqi in control groups.The parietal cells were isolated and purified with Gastric Sac Method described by Lewin and Percoll density gradient separation.Radioligand binding assay was established in screening gastrin receptor of parietal cells.RESULTS:The symptoms of the rats in spleen deficiency model groups were similar to which of the patients in clinic.These symptoms were markedly ameliorated after treated by Huangqi. In spleen deficiency model rats caused by reserpine,the gastrin receptor bnding sites of parietal cells decreased markedly(271.2±91.8).Which were lower than that in normal group (619.1±304.5, P<0.05 ).Binding sites were increased remarkably by administration of Huangqi (545.2±205.3, P<0.01 ).CONCLUSION:That Gas R was in a lower metabolic condition in spleen deficiency rats.And it could be remarkably increased by invigorating spleen and replenishing Qi herbs.There is a close relationship between spleen deficiency and gastrin receptor.
