糖尿病周围神经病变患者血清脑源性神经营养因子与高迁移率族蛋白B1水平相关性的研究 被引量:11

Association between the serum BDNF and HMGB1 levels in T2DM patients with peripheral neuropathy

摘要 目的探讨糖尿病周围神经病变(DPN)患者血清脑源性神经营养因子(BDNF)与高迁移率族蛋白B1(HMGB1)水平的相关性。方法将T2DM患者60例分为T2DM组和DPN组,每组各30例,另选取健康体检者30名作为对照(NC)组。ELISA检测血清BDNF及HMGB1含量,RT-PCR法检测其mRNA在外周血单个核细胞中的相对表达水平,分析DPN患者中BDNF与HMGB1相关性。结果DPN组和T2DM组血清BDNF含量及mRNA表达水平均低于NC组(P<0.05),而HMGB1含量及mRNA表达水平高于NC组(P<0.05)。DPN组血清BDNF含量及mRNA表达水平低于T2DM组,血清HMGB1含量及mRNA表达水平高于T2DM组(P<0.05)。DPN组血清BDNF与HMGB1水平负相关(r=-0.425,P<0.05)。结论DPN患者血清BDNF与HMGB1相关,二者可能共同参与了DPN的发病。 Objective To investigate the association between serum brain-derived neurotrophic factor (BDNF) and high-mobility group box-1 (HMGB1) levels in T2DM patients with peripheral neuropathy. Methods Sixtiy patients with type 2 diabetes mellitus were divided into T2DM group (n= 30) and DPN group (n= 30). 30 healthy subjects were collected as normal control (NC) group. The serum levels of BDNF and HMGB1 were tested by ELISA, BDNF and HMGB1 mRNA were detected by quantitative RT-PCR in peripheral blood mononuclear ceils. The correlation analysis was carried out between BDNF and HMGB1. Results The serum level of BDNF and the expression level of mRNA were lower in T2DM and DPN groups than in NC group (P^0.05),whereas the serum level of HMGB1 and its mRNA expression was higher. The serum level of BDNF and its mRNA expression level of mRNA were lower, but the serum level of HMGB1 and expression level of mRNA was higher in DPN group than in T2DM group(P〈0.05). In DPN group, correlation analysis showed a significant relationship between BDNF and HMGBI(r=-0. 425,P〈0.05). Conclusion There is a significant correlation between the levels of HMGB1 and BDNF in type 2 diabetic patients with peripheral neuropathy. Both BDNF and HMGB1 may participate in the process of type 2 diabetic patients with peripheral neuropathy.
