摘要
目的:研究参麦注射液(shenmai injection,SMI)对大鼠H9c2心肌细胞中细胞色素P450(cytochrome P450,CYP450)m RNA表达的影响。方法:通过传代的方法培养心肌细胞,通过细胞计数试剂盒(cell counting kit-8,CCK-8)检测给药后细胞的活性,乳酸脱氢酶(lactate dehydrogenase,LDH)实验检测细胞毒性,最终选取合适的浓度和给药时间对细胞进行处理,采用realtime PCR方法,检测不同给药浓度下各处理组对大鼠H9c2心肌细胞中CYP450亚酶CYP2C11、CYP2J3、心房利钠肽(atrial natriuretic polypeptide,ANP)和脑钠肽(brain natriuretic peptide,BNP)m RNA表达的影响。结果:SMI对H9c2细胞CYP2J3和CYP2C11的表达具有诱导的作用,在ANP和BNP的m RNA表达水平上,SMI H9c2细胞也具有诱导的作用,且随着浓度的增加,诱导作用逐渐增强。结论:CYP2J3、CYP2C11、ANP和BNP等相关分子是心血管的重要靶点,而SMI对心血管疾病疗效明确,因此心脏P450酶系统可能是SMI治疗心血管疾病新的途径或靶点。
Objective:To investigate the influence of Shenmai injection(SMI) on mRNA expression of cytochrome P450 (CYP450) in H9c2 myocardial cell. Methods:H9c2 cell was cultured by method of passage. Cell counting kit-8(CCK-8) assay was used to detect the viability of H9c2 cells and lactate dehydrogenase(LDH) assay was used to detect the toxicity of H9c2 cells. The expression of CYP 2J3,CYP2C11,atrial natriuretic polypeptide(ANP) and brain natriuretic peptide(BNP) were measured by real-time PCR. Results:SMI could induce the expression of CYP 2J3 and CYP2Cll in H9c2 cell. In addition,SMI could induce the mRNA of ANP and BNP in a dose-dependent manner. Conclusion:SMI was widely used in the treatment of cardiovascular diseases. CYP2J3, CYP2C11, ANP and BNP mRNA expression maybe the new targets of metabolism.
