摘要 目的探讨赤芍与菟丝子对豚鼠皮肤黑素细胞酪氨酸酶活性及mRNA基因表达水平的影响,为应用中药治疗白癜风等色素减退性皮肤病提供实验依据。方法利用棕黄色豚鼠背部皮肤色素实验动物模型,多巴氧化法测定酪氨酸酶活性,然后采用半定量逆转录聚合酶链反应(RT-PCR)检测药物干预前后豚鼠皮肤中酪氨酸酶及相关蛋白-1 mRNA表达水平的变化。结果赤芍与菟丝子均呈剂量依赖性地促进酪氨酸酶活性的增加;与正常对照组相比,1g/(kg·m L)的赤芍、菟丝子组酪氨酸酶及相关蛋白-1mRNA表达水平上升,且差异有统计学意义(P<0.01);与阳性对照组8-MOP相比差异无统计学意义(P>0.05)。结论赤芍、菟丝子的乙醇提取液均可上调豚鼠皮肤黑素细胞酪氨酸酶活性及促进mRNA表达,促进皮肤黑素生成。 Objective To investigate the effect of Paeonia veitchii lynch and Cuscuta chinensis lam on tyrosinase activi- ty and expression of mRNA in skin of guinea pigs, and provide the experimental evidence for treating pigmental disorders such as vitiligo. Methods Experiments were done using the pigmental back skin of brown guinea pig. Tyrosinase activity was detected by dopa oxidase assay. Tyrosinase and TRP-1 mRNA expression was evaluated by RT-PCR before and after drug intervention. Results Paeonia veitchii lynch and Cuscuta chinensis lam could upregulate tyrosinase activity in a dose-dependent manner. In the lg/( kg · mL) group, tyrosinase and TRP-1 mRNA expression elevated, showing significant difference ( P 〈 0.01 ) as compared to the normal control group. No significant difference ( P 〉 0.05 ) were observed when compared to the positive control group of 8-MOP. Conclusion Ethanol extracts of Paeonia veitchii lynch and Cuscuta chinensis lam can up-regulate melanocytes tyrosinase activity and mRNA expression, and then induce the generation of cutaneous melanin.
