摘要
目的:探讨黑蒜提取液对结肠癌Ht-29移植瘤辐射增敏作用及可能机制。方法:建立结肠癌Ht-29皮下移植瘤模型,对4组移植瘤模型分别给予0.9%氯化钠溶液(对照组)、60Coγ射线照射(照射组)、黑蒜提取液(黑蒜组)及黑蒜提取液联合60Coγ射线照射(黑蒜+照射组)的处理,实验结束后摘取眼球取血后脱臼处死小鼠。剥离瘤体,计算肿瘤生长抑制率;ELISA法检测血清中血管内皮生长因子(VEGF);免疫组织化学法检测肿瘤组织中丝氨酸苏氨酸激酶(Akt)蛋白的表达变化;蛋白质印迹法检测Akt、p Akt蛋白的表达。结果:与对照组、照射组、黑蒜组相比,黑蒜提取液联合60Coγ射线可显著抑制结肠癌Ht-29移植瘤的生长,降低VEGF、Akt及p Akt蛋白表达。结论:黑蒜提取液能够增加结肠癌Ht-29移植瘤辐射敏感性,其作用机制可能与下调Akt、p Akt蛋白表达及VEGF从而降低其放射抵抗有关。
Objective: It explored the radiosensitization effect and possible mechanism on colon cancer Ht-29 transplanted tumors with black garlic extract. Methods: The tumor-bearing C57BL/6 mouse model was established, 40 mice were randomly divided into 4 groups: control group, irradiation group, black garlic group, black garlic and irradiation group. After the experiment, mice were sacrificed. Then the tumors were separated and weighed to calculate the tumor-inhibiting rate. The ELISA method was used to detect serum vascular endothelial growth factor (VEGF). AKt protein expression was examined by Immunohistochemistry method. The protein expressions of Akt and pAkt were examined by Western blot. Results: Compared with the control group, irradiation group and black garlic group, black garlic extract combined radiotherapy could increase the tumor-inhibiting rate and reduce the protein expression of VEGF, Akt and pAkt significantly. Conclusion: Black garlic extract could enhance the radiation sensitivity of colon cancer Ht-29 transplantation tumors. The action mechanism may be associated with down-regulation of VEGF, Akt and pAkt protein expression.
