摘要
目的:探讨银杏总黄酮对异丙基肾上腺素致心力衰竭大鼠的心功能和血管紧张素Ⅱ(AngⅡ)、血清内皮素1(ET-1)的影响。方法:选用SPF级雄性SD大鼠40只,随机分为4组:空白组、模型组、银杏总黄酮组和美托洛尔组。除空白组外均给予皮下注射300mg/kg异丙基肾上腺素2次,造模成功后银杏总黄酮组给予200mg·kg-1·d-1的银杏总黄酮提取物,美托洛尔组给予美托洛尔20mg/kg,其余灌胃给予相同体积的0.9%氯化钠溶液,连续8周。实验结束后测定大鼠心功能,血清内ET1和AngⅡ含量和病理积分,并取心肌观察病理形态学特征。结果:银杏总黄酮组和美托洛尔组左心室射血分数、室间隔厚度和左心室后壁厚度降低,左心室舒张末内径、左心室收缩末内径升高,与模型组有显著差异(P<0.05),表明治疗后心功能有所提高;银杏总黄酮组ET-1、AngⅡ含量和病理评分明显下降,与模型组相比差异有统计学意义(P<0.05)。结论:银杏总黄酮可改善心力衰竭大鼠的心功能,其作用机制可能与降低ET-1、AngⅡ的含量有关。
Objective: To explore the effects of the total flavone of ginkgo (TFG) on cardiac function, Ang Ⅱ and ET-1 of rats with heart failure induced by isopropyl adrenaline. Methods: 40 male SPF Sprague-Dawley rats were randomly divided into 4 groups: blank group, model group, TFG group and metoprolol group. Expect for the blank group, all rats were received subcutaneous injection of isopropyl adrenaline at 300mg/kg, for twice. After the model was successfully established, the TFG group and metoprolol group were respectively given TFG extract at 200mg·k^g-1.d^-1 and the metoprolol at 20mg/kg, while the other groups were garaged the same volume of normal saline, lasting for 8 weeks. After the experiment, the level of ET-1, the content of Ang Ⅱ in serum and the pathological scores were detected, and the pathologic morphological characteristics of myocardial tissue were determined. Results: Compared with those in the model group, left ventricular ejection fraction (LVEF), interventficular septum thickness (LVET) and left ventricular posterior wall thickness (LVPWT) were significantly decreased and left ventricular end diastolic diameter (LVEDD) and left ventricular end systolic dimension (LVESD) were increased in TFG group and metoprolol group, with significant differences between the TFG group, metoprolol group and the model group (P〈0.05). It suggested that cardiac function was improved after the treatment of TFG or metoprolol. The level of ET-1, the content of Ang Ⅱ in serum and the pathological scores in TFG group were lower than those in the model group, with significant differences between the TFG group and the model group (P〈0.05). Conclusion: Ginkgo flavonoids extract can improve cardiac function of rats with heart failure, the mechanism of which may be related to the levels of ET-1 and Ang Ⅱ.
