摘要
本文以垂盆草的茎段为主要试验材料,测定了适合垂盆草生长的各种培养基中的最适浓度,包括蔗糖、BA、NAA;此外对垂盆草组织培养高效再生体系的建立进行了探索,得到的主要研究结果如下:(1)研究垂盆草的培养基类型,得出1/2MS基本培养基最适合垂盆草的生长。(2)研究含有1%、2%、3%、4%、5%蔗糖浓度的培养基,发现3%的蔗糖浓度最适合垂盆草的生长;(3)研究含有0.2、0.4、0.6、0.8mg/L的NAA浓度的培养基,发现0.4mg/L的NAA浓度最适合垂盆草的生长;同样得出0.2mg/L的BA浓度最适合垂盆草的生长。(4)适合垂盆草组织培养高效再生体系的基本培养基配方为:1/2MS+0.4mg/L NAA+0.2mg/L BA+3%蔗糖+0.7%琼脂的培养基。同时观察试验结果时,可以看出:适当的增减BA的浓度,可以使茎秆粗壮。
In this thesis sarmentosum stem segments as the main test material were determined suitable sarmentosum growth of various optimum concentration of the medium, including sucrose, BA, NAA ; Also on sarmento sum tissue culture regeneration system establishment are explored to obtain the main resuhs are as follows: (1) Study sarmentosum medium type, draw 1/2MS basic medium best suited sarmentosum growth.
(2)Study containing 1%, 2%, 3%, 4%, 5% sucrose medium, 3% sucrose concentration found most suitable for the growth of sarmentosum ;
( 3 ) The study contains 0. 2,0. 4,0. 6,0. 8mg / L of NAA concentrations of medium and found 0.4mg / L NAA concentration of the most suitable sarmentosurn growth ; equally drawn 0.2mg / L of BA was the most suitable vertical pots of grass growth.
(4) Suitable sarmentosum tissue culture regeneration system of the basic medium for: 1/2MS + 0.4mg/L NAA + 0.2mg / L BA + 3% sucrose + 0.7% agar medium. While observing the test results, it can be seen : the appropriate changes in the concentration of BA, can make thick stems.
