邻苯二甲酸二辛酯和氯仿联合染毒对雄性小鼠脑组织脂质过氧化及能量代谢的影响 被引量:3

Effects of dioctylphthalate and chloroform exposure in combination on lipid peroxidation and energy metabolism in brain tissue of male mice

摘要 目的观察邻苯二甲酸二辛酯(dioctylphthalate,DOP)和氯仿(chloroform,CF)联合染毒对雄性小鼠脑的脂质过氧化及能量代谢的影响。方法将20只成年清洁级ICR雄性小鼠按照2×2析因设计随机分为4组,分别为对照(玉米油)组、DOP单独染毒组(1/10 LD50,540 mg/kg)、CF单独染毒组(1/10 LD50,112 mg/kg)和DOP(1/10 LD50,540 mg/kg)+CF(1/10LD50,112 mg/kg)联合染毒组,每组5只。灌胃染毒,连续15 d。测定小鼠脑组织中丙二醛(MDA)、一氧化氮(NO)含量、钙离子浓度和过氧化氢酶(CAT)、乳酸脱氢酶(LDH)、琥珀酸脱氢酶(SDH)、一氧化氮合酶(NOS)、ATP酶活力及线粒体膜电位。结果与对照组比较,仅CF单独染毒组小鼠脑组织中MDA的含量较高,CF单独染毒组及DOP+CF联合染毒组小鼠脑组织中Na+-K+-ATP酶、Ca2+-Mg2+-ATP酶、NOS活力和线粒体膜电位升高,DOP单独染毒组及DOP+CF联合染毒组小鼠脑组织中LDH、SDH活力和NO含量降低,差异均有统计学意义(P<0.05);三组小鼠脑组织中CAT的活力及钙离子浓度均无明显改变。DOP+CF联合染毒对小鼠脑组织中的线粒体膜电位表现协同作用,Na+-K+-ATP酶活力表现为增强作用,MDA含量和LDH、SDH、Ca2+-Mg2+-ATP酶活力表现拮抗作用,而NOS活力、NO含量尚未表现出联合作用。结论DOP和CF联合染毒尚未造成小鼠脑组织脂质过氧化损伤,可促进其能量代谢。 Objective To study the effects of dioctylphthalate(DOP) and chloroform(CF) exposure in combination on lipid peroxidation and energy metabolism in the brain tissue of male mice. Methods ICR mice were randomly divided into control group(corn oil),DOP exposure group(1/10 LD50,540 mg/kg),CF exposure group(1/10 LD50,112 mg/kg) and DOP(1/10LD50,540 mg/kg)+CF(1/10 LD50,112 mg/kg) exposure groups, the treatment were conducted through gavage, and the mice were exposed every day for 15 consecutive days. The brain tissue was collected and the contents of MDA,NO,Ca2+,the activities of CAT,LDH,SDH,NOS,ATPase and mitochondrial membrane potential were examined. Results Compared with control group,only the content of MDA in chloroform group increased significantly(P 〈0.05),the activities of Na+-K+-ATPase,Ca2 +-Mg2 +-ATPase,NOS and the mitochondrial membrane potential in chloroform group and combined exposure group increased significantly(P〈0.05),the activities of LDH,SDH and the content of NO in DOP group and combined exposure group reduced significantly(P〈0.05). The activity of CAT and the concentration of Ca2+did not exist combined effect in all the groups. In the combined exposure group,the mitochondrial membrane potential existed synergistic effect,the activity of Na+-K+-ATPase existed potentiation,the content of MDA,the activities of LDH,SDH,Ca2+-Mg2+-ATPase existed antagonism,the activity of NOS and the content of NO did not exist combined effect. Conclusion DOP and CF in combination can not produce lipid peroxidation injury and may accelerate energy metabolism in male mice brain.
