摘要
目的探讨斑马鱼急性毒性检测平台在食品安全性检测中的应用性。方法通过完善和优化斑马鱼驯养条件、斑马鱼急性试验用鱼要求、斑马鱼急性毒性试验条件建立斑马鱼急性毒性检测平台,检测和分析不同浓度的硫氰酸钠、蛹虫草子实体水提物和醇提物对斑马鱼的毒性作用,分别观察和记录斑马鱼在受试物的试验溶液中暴露48h的毒性反应和死亡情况。结果斑马鱼在剂量为1000 mg/L和800 mg/L的硫氰酸钠试验溶液饲养1 h后开始出现毒性反应,在剂量为600 mg/L和400 mg/L的硫氰酸钠试验溶液饲养4h后开始出现毒性反应,硫氰酸钠对斑马鱼的48h LC5 0为572.4 mg/L。斑马鱼在浓度为100-1 000 mg/L的蛹虫草子实体水提物试验溶液中饲养48h,未发现斑马鱼有任何毒性反应,蛹虫草子实体水提物对斑马鱼的48h LC5 0大于1 000 mg/L;斑马鱼在浓度为100-1 000 mg/L的蛹虫草子实体醇提物试验溶液中饲养48h也未发现斑马鱼有任何毒性反应,蛹虫草子实体醇提物对斑马鱼的48h LC5 0大于1000mg/L。结论斑马鱼急性毒性检测平台能够用于食品和食品污染物急性毒性的快速检测,该检测平台将在食品的安全性检测中有较好的应用前景。
Objective To discuss the application prospect of zebrafish toxicity testing platform used in food safety test. Methods Through the improvement and optimization of zebrafish domestic condition,zebrafish acute test fish requirements,zebrafish acute toxicity test conditions,established zebrafish acute toxicity testing platform,to detect and analyze the toxic effect of sodium sulfocyanate,cordyceps militaris fruiting body water extract and alcohol extract with different concentrations on zebrafish,toxicity reaction and death situation were observed and recorded for zebrafish been exposed in test solution of test samples for 48 h. Results The toxic reaction of zebrafish began to appear after zebrafish were feed in the test solution containing sodium sulfocyanate of 1000 mg / L and 800 mg / L for 1h,toxic reaction of zebrafish began to appear after zebrafish were feed in the test solution containing sodium sulfocyanate of 600 mg / L and 400 mg / L for 4h,the LC5 0of sodium sulfocyanate in48 h was 572. 4 mg / L. Zebrafish were feed in the test solution containing cordyceps militaris fruiting body water extract for 100-1000 mg / L for 48 h,no toxicity was found,the LC5 0of cordyceps militaris fruiting body water extract in 48 h is greater than 1000 mg / L; Zebrafish were feed in the test solution containing 10-1000 mg / L cordyceps militaris fruiting body alcohol extract for 48 h,no toxicity was found neither,the LC5 0of cordyceps militaris fruiting body alcohol extract in 48 h is greater than 1000 mg / L. Conclusions Acute toxicity of zebrafish testing platform can be used as rapid detection method for the acute toxicity of food and food contaminants,so it will be wide applied to the food safety test.
