摘要
目的探究复方丹参滴丸对大鼠视网膜缺血再灌注损伤(retinal ischemia reperfusion injury,RIRI)的影响。方法将42只健康成年SD大鼠随机分为正常对照组(6只)、模型组(18只)和治疗组(18只)。模型组和治疗组采用前房灌注加压建立大鼠RIRI模型,治疗组RIRI后立即给予复方丹参滴丸(450 mg·kg-1·d-1)灌胃,连续7 d,模型组和正常对照组灌胃等量生理盐水。观察RIRI后24 h、7 d视网膜电流图的改变,并取视网膜行组织病理学观察。测量RIRI后24 h、7 d视网膜各层厚度,同时计数视网膜神经节细胞数。结果 RIRI后24 h及7 d,模型组视网膜电流图的b波振幅分别为(92.33±24.36)μV及(101.28±21.97)μV,较正常对照组大鼠测量到的(215.40±35.91)μV显著降低,而治疗组的b波振幅分别为(137.07±22.71)μV及(166.24±16.72)μV,较模型组显著好转(均为P<0.05);各组a波与b波的变化趋势相同。大鼠RIRI后24 h,模型组全层视网膜高度水肿,可见大量空泡变性,RIRI后7 d,视网膜厚度明显变薄,而治疗组大鼠视网膜的组织病理学损害较模型组均明显减轻。结论复方丹参滴丸可从形态学及功能学上减轻大鼠RIRI损伤。
Objective To study the effects of compound danshen dripping pills on retinal ischemia-reperfusion injury( RIRI) in rats. Methods Forty-two adult healthy SD rats were randomly divided into 3 groups: Sham group( 6 rats),model group( 18 rats) and treatment group( 450 mg·kg^-1·d^-1)( 18 rats). The model of RIRI was established by increasing the intraocular pressure via cannulation into the anterior chamber. Electroretinogram( ERG) was recorded at24 hours and 7 days after RIRI,respectively,then pathological changes of the retina were observed with light microscope. Thicknesses of the retinal layers and RGC cell counts were calculated at 24 hours and 7 days respectively after RIRI. Results At 24 hours and 7 days after RIRI,the b-wave amplitudes of the model group were( 92. 33 ± 24. 36) μV and( 101. 28 ± 21. 97) μV,respectively,which were much lower than the sham group( 215. 40 ± 35. 91) μV. While the bwave amplitudes of the treatment group were( 137. 07 ± 22. 71) μV and( 166. 24 ± 16. 72) μV,respectively,which were higher than the model group( all P〈0. 05). In the model group,all layers of the retinas were highly edema with enormous vacuolar degeneration at 24 hours after RIRI,and the thicknesses of retinas were greatly reduced at 7 days after RIRI,as shown in the HE analysis. These insults were reversed to some extent in the compound danshen dripping pills treatment group. Conclusion Compound danshen dripping pills may have a protective effect on the retinal morphology as well as the retinal function after RIRI.
