重离子辐照对人舌鳞癌Tca8113细胞细胞周期及VEGFmRNA的影响 被引量:2

Effects of Carbon Ions on Cell Cycle and the Expression of VEGF in Human Tongue Squamous Carcinoma Tca8113 Cell Line

摘要 目的:观察重离子对人舌鳞癌Tca8113细胞细胞周期及VEGFmRNA表达的影响。方法:分别用0、1、4Gy的重离子及X线辐照体外培养的人舌鳞癌Tca8ll3细胞,流式细胞技术观察细胞周期的变化;原位杂交法检测VEGFmRNA表达。结果:流式细胞术结果,OGy组未出现阻滞,重离子照射组剂量1Gy时,12h出现G2/M期阻滞,4Gy时在36、48h出现G2/M期明显阻滞,X线照射组在1Gy时未出现阻滞,4Gy时,24、36h出现G2/M期阻滞,与0Gy组相比,差异有统计学意义(P<0.05)。原位杂交结果,与0Gy组相比,1Gy时经重离子及X线辐照后Tca8113细胞VEGFmRNA表达降低,4Gy时,重离子组VEGFmRNA表达明显降低,X线组4Gy辐照后VEGFmRNA阳性表达率与重离子组1Gy差异无统计学意义。结论:重离子较X线对舌鳞癌细胞细胞周期及VEGFmRNA影响更明显,有更可靠的抑癌效果。 Objective:To observe the effects of carbon ions on cell cycle and the expression of vascular endothelial growth factor mRNA(VEGFmRNA)in human tongue squamous carcinoma Tca8113 cell line.Methods:Tca8113cells were irradiated with carbon ions and X-rays with the doses of 0Gy,1Gy,and 4Gy respectively.The untreated control and irradiated cells were observed by flow cytometry(FCM)and in-situ hybridyzation.Results:Results showed the cells were arrested in G2/M phase 12 hours post-irradiation by carbon ions with the dose of 1Gy,while it was more obvious after 36 hours and 48 hours with the radiation dose of 4Gy.The cells irradiated by X-rays with the dose of 1Gy were not arrested at G2/M checkpoint.In contrast,the G2/M arrest appeared at the 24 and 36hours after irradiation by 4Gy.Compared with the control group(0Gy),the difference was statistically significant(P〈0.05).The results obtained from in situ hybridization showed that after carbon ions and X-rays radiation of 1Gy,VEGFmRNA expression was reduced compared with 0Gy group.Moreover,the expression of VEGFmRNA was decreased markedly in the group of carbon ions radiation with 4Gy,and the expression of VEGFmRNA induced by X-rays was equal with that of carbon ions group with 1Gy.Conclusion:Compared with X-rays,carbon ions is more effective in inhibiting the cancer cells in vitro.
