长双歧杆菌抑制回肠NLRP6炎症小体调节感染后肠易激综合征内脏高敏感性 被引量:7

Bifidobacterium longum regulates the visceral hypersensitivity of PI-IBS by inhibiting NLRP6 inflammasome

摘要 [目的]研究NLRP6炎症小体在长双歧杆菌调节感染后肠易激综合征(PI-IBS)内脏高敏感性中的作用。[方法]NIH小鼠40只分为5组(每组8只):2周正常对照组(NC-2W组),感染后2周组(PI-2W组),8周正常对照组(NC-8W组),感染后8周组(PI-8W组)及感染后8周长双歧杆菌干预组(PI-B组);腹壁回撤反射(AWR)检测各组内脏敏感性,Western-blot检测CD172a以评价肠道炎症程度,Western-blot检测各组小鼠末端回肠NLRP6及其配体ASC、caspase-1、CARD8以及下游细胞因子白细胞介素(IL)-18、IL-1β表达。[结果]1PI-2W组和PI-8W组内脏敏感性较NC-2W组和NC-8W组明显增加(P<0.05),PI-B组内脏敏感性较PI-8W组明显降低(P<0.05);2PI-2W组与NC-2W组比较,CD172a表达水平显著升高(P<0.01);PI-8W组与NC-8W组比较,CD172a表达水平差异无统计学意义(P>0.05);PI-B组与PI-8W组比较,CD172a表达水平差异无统计学意义(P>0.05);3 PI-2W组和PI-8W组NLRP6、CARD8、ASC和caspase-1及IL-18均较NC-2W组和NC-8W组明显增加(P<0.05),而PI-B组NLRP6、CARD8、ASC和caspase-1及IL-18较PI-8W组明显降低(P<0.05),PI-2W组IL-1β表达水平较NC-2W组显著升高(P<0.05),而NC-2W组、NC-8W组、PI-8W组、PI-B组间IL-1β表达水平比较均差异无统计学意义(P>0.05)。[结论]长双歧杆菌可能通过抑制炎症小体NLRP6,下调IL-18,从而降低PI-IBS内脏高敏感性。 [Objective]To investigate the role of NLRP6 inflammasome in Bifidobacterium longum regu-lating the visceral hypersensitivity of PI-IBS.[Methods]Forty NIH mice were randomly divided into five groups:normal controls of 2 weeks(NC-2W group) ,2 weeks after infection (PI-2W group) ,normal control of 8 weeks(NC-8W group) ,8 weeks after infection group(PI-8W group)and Bifidobacterium longum inter-vention group(PI-B group).Visceral sensation evaluation of abdominal withdrawal reflex score was per-formed for all mice before they were sacrificed.The inflammation degree was evaluated by CD172 a in dif-ferent groups.The terminal ileum tissue of each mouse was removed for detecting the expression of NL-RP6 ,ASC ,CARD8 ,caspase-1 by Western-blot.IL-18 and IL-1βexpression were further tested by Western-blot.[Results]The visceral sensitivity of PI-2W group and PI-8W group increased significantly compared with their control groups(P〈0.05) ,however ,the visceral sensitivity of PI-B group had a obvious decline&amp;nbsp;compared with PI-8W group(P〈0.05).The expression of CD172 a in PI-2 W group increased significantly compared with NC-2 W group(P〈0.05) ,however ,there was no significant difference of CD172 a expres-sion in PI-8W group ,NC-8W group and PI-B group(P〉0.05).The expression of NLRP6、CARD8、ASC、caspase-1 and IL-18 in PI-2 W and PI-8W group increased significantly when compared with their control groups(P〈 0.05).The expression of NLRP6 ,CARD8 ,ASC and caspase-1 in PI-B group decreased signifi-cantly when compared with PI-8W group(P〈 0.05).IL-1βexpression increased remarkably only in PI-2 W group ,however ,there were no obvious difference among control group ,PI-8W group and PI-B group(P〉0.05) .[Conclusions]Bifidobacterium longum decreased the visceral hypersensitivity of PI-IBS through in-hibiting the expression of NLRP6 and further downregulated IL-18 expression.
