摘要
目的观察卵白蛋白(OVA)致大鼠过敏抗血清对不同品系小鼠被动皮肤过敏试验(PCA)及血液免疫球蛋白E(IgE)和免疫球蛋白G(IgG)抗体水平变化的影响。方法 Wistar大鼠随机分为生理盐水(NS)组、NS佐剂组、OVA组、OVA佐剂组,采用皮下注射方式给予相应药物致敏,制备大鼠OVA抗体血清,并分别将其注入不同品系小鼠耳廓内,于致敏后3 h或48 h以相应药物激发,观察各组小鼠耳廓蓝染情况,并用酶联免疫吸附剂测定(ELISA)检测小鼠体内抗体水平变化。结果各品系小鼠(KM、BALB/c、ICR、C57BL/6、B6;129S-Fcgr2btm1Ttk/J)NS组、NS佐剂组、OVA组耳廓蓝染阴性,OVA佐剂组耳廓蓝染阳性,其中各品系间存在差异,小鼠耳异种PCA敏感程度强度依次为KM、BALB/c>ICR、C57BL/6、B6;129S-Fcgr2btm1Ttk/J。结论小鼠异种PCA试验中,不同品系动物对OVA抗血清的敏感性存在差异,其中KM,BALB/c小鼠较为敏感,但PCA强度与血液IgE水平无明显的相关性。
AIM To compare the differences among ICR,C57BL/6,BALB/c,KM and B6; 129S-Fcgr2btm1Ttk/J mice in passive cutaneous anaphylaxis test by Wistar rat serum against ovalbumin (OVA) and the influence on mice serum IgE and IgG.METHODS Wistar rats were randomly divided into 4 groups (normal saline (NS),NS + alum,OVA,OVA + alum),and each group sensitized by OVA through subcutaneous injection.Serum samples were taken 11 days later,and then had a xenogenic PCA experiment between rats and mice,3 or 48 hours later stimulated with relative drugs.The situation of locus coeruleus was observed and the mice serum antibodies were assayed by ELISA method.RESULTS The group of OVA + alum was positive in xenogenic PCA experiment among all strains of mice,while other groups were negative.There was some difference among those strains,and the sensitivity to rat OVA antiserum of mice was KM,BALB/c > ICR,C57BL/6,C2848.CONCLUSION In the xenogenic PCA experiment between rats and mice,there is detectable difference in sensitivity among those mice strains with KM and BALB/c mice being more sensitive.There is no obvious relevancy between the intensity of PCA and antibodies in mice serum.
