摘要：目的：建立一种快速、灵敏的正向高效液相色谱法测定角鲨烯胶囊中角鲨烯的含量。方法：角鲨烯胶囊内容物经异辛烷溶解稀释后，采用SunFire^TM Prep Silica色谱柱、0．5％甲醇-0．5％正辛醇-异辛烷流动相进行分离，紫外检测器210nm处进行检测。结果：10～1000μg／mL浓度范围内角鲨烯呈良好的线性关系，相关系数r=0．999，最低定量限为10μg／mL．低、中、高3个浓度水平的加标回收率均值为102．1％，相对标准偏差为1．0％。采用所建方法对市售3批角鲨烯胶囊进行含量测定，角鲨烯标示含量均符合规定。结论：所建方法适合角鲨烯胶囊中角鲨烯的含量测定。
Objective：To establish a rapid and sensitive method for determination ofsqualene in squalene capsules by normal-phase high performance liquid chromatography. Methods：Squalene was extracted and was dissolved and diluted by isooctane, using a mobile phase of isooctane with 0.5% methanol and 0.5% n-octyl alcohol on SunFire TM Prep silica column by UV-detector at 210 nm. Results：The calibration curves showed good linear.relationship in the range of 10 - 1000μg/mL for squalene with the correlation coefficient of 0.999.The limit of quantification （LQD） of squalene wasl0μg/mL. The average recovery of squalene was 102.1% and the relative standard deviations was 1.0 %. Three batches ofsqualene capsules were measured by our method, which showed that the content ofsqualene in all tested samples met the specified requirements. Conclusion：The method was proved to be suitable for determination ofsqualene in squalene capsules.
