D-半乳糖和氢化可的松诱导老年阳虚证大鼠模型的建立 被引量:6

Establishment of Aging Rat Model with Yang-deficiency by D-galactose and Hydrocortisone

摘要 目的探讨如何利用青年大鼠建立合适的老年阳虚证模型。方法选用3月龄SD健康大鼠30只,雌雄各半,随机分为青年对照组,亚急性衰老模型组与老年阳虚证模型组,每组10只。亚急性衰老模型组给予颈背部皮下注射D-半乳糖125mg/kg,连续40d,之后于后肢肌肉注射25mg/kg生理盐水7d;老年阳虚证模型组给予颈背部皮下注射D-半乳糖125mg/kg,连续40d,后再行后肢肌肉注射氢化可的松25mg/kg,连续7d;青年对照组在颈背部皮下及后肢肌肉注射等量生理盐水47d。检测大鼠血清中IL-6、IL-2、cAMP、cGMP和CD4+CD8+T淋巴细胞的含量。结果与青年对照组相比,亚急性衰老组大鼠cGMP水平升高(P<0.05),cAMP水平及cAMP/cGMP比值无明显变化(P>0.05);而老年阳虚组大鼠cGMP明显升高(P<0.01),cAMP水平及cAMP/cGMP比值明显降低(P<0.01)。亚急性衰老组和老年阳虚组大鼠血清中IL-6含量均明显上升,IL-2含量均明显下降(P<0.05或P<0.01);CD4+T淋巴细胞比率降低,CD8+T淋巴细胞比率升高,CD4+/CD8+比值下降(P<0.01或P<0.05)。与亚急性衰老组相比,老年阳虚组大鼠cAMP明显下降,cGMP明显上升,cAMP/cGMP比值下降(P<0.01);血清中IL-6、IL-2含量以及CD4+、CD8+T淋巴细胞比率以和CD4+/CD8+比值较亚急性衰老组又均有明显差异(P<0.05)。结论以D-半乳糖和氢化可的松诱导制作的老年阳虚证模型大鼠符合衰老及阳虚的表现,提示以此种方法制作老年阳虚证模型具有可行性。 Objective To explore how to make use of the young rats establish proper aging model rats with yang - deficiency. Methods 30 healthy SD rats,3 months old, were randomly divided into three groups:young control group,Subacute aging model group, aging with yang - deficiency model group. Every rats in Subacute aging model group were injected with D - galactose subcutaneously for 40 days to induce the sub - acute aging model rats, then all rats were injected with saline for 7 days. Every rats in aging with yang - deficiency model group were injected with D - galactose subcutaneously for 40 days, then all rats were injected with hydrocortisone for 7 days to establish the yang - deficiency symptom on the aging model rats. The rats in young control group were injected of same amount of saline for 47 days. Detect the content of cAMP,cGMP,IL - 6 ,IL - 2 and the content of CIM + CD8 + T lymphocytes in the serum of rats. Results In comparison with the young control group ,the level of cGMP in Subacute aging model group inereased(P 〈 0.05), but the level of cAMP and the ratio of cAMP/cGMP has no obvious change (P 〉 O. 05). However, the level of cGMP raised remarkably, while the level of cAMP and the ratio of cAMP/cGMP declined remarkably in aging with yang - deficiency model group( P 〈 0.01 ). In the above two groups ,the content of IL - 6 and the ratio of CD8 + T lymphocytesbut increased, the content of IL - 2 declined (P 〈 0.05 或 P 〈 0.01 ) ; and the ratio of CD4 + T and CIM + / CD8 + T lymphocytes decreased( P 〈 0.05 或 P 〈 0.01 ). In comparison with the Subacute aging model group, the level of cGMP raised re- markably, and the level of cAMP and the ratio of cAMP/cGMP declined remarkably in aging with yang - deficiency model group ( P 〈 0.01 ).Meanwhile the content of IL - 6 ,IL - 2 and the ratio of CD8 + CD4 + ,CD4 +/CD8 + T lymphocytesbut have significant differences ( P 〈 0.05 ).
