81种药用植物提取物影响小鼠巨噬细胞RAW264.7活化的研究

摘要 目的通过对81种药用植物提取物影响巨噬细胞活化的研究,获得相关活性数据,为后续研究开发和利用提供试验数据和物质基础。方法将提取物样品作用于RAW264.7细胞一定时间后,用ELISA法测定细胞分泌的TNF-α,以判断样品对细胞是否有活化作用;将LPS和81种提取物样品作用于RAW264.7细胞一定时间后,用ELISA法测定细胞分泌的TNF-α,以判断样品是否对LPS诱导的细胞活化有抑制作用。结果对81种药用植物提取物的研究结果表明,有34种植物提取物样品能够使RAW264.7细胞TNF-α表达上调,使其增加TNF-α的分泌;有2种植物提取物样品能够使LPS诱导的RAW264.7细胞TNF-α表达下调,使其减少TNF-α的分泌。结论通过对81种药用植物提取物的筛选研究表明,既有对巨噬细胞有活化作用的样品,也有能够抑制LPS诱导的巨噬细胞活化的样品,为进一步的研究,提供了相关活性数据和物质基础。 Aim To provide scientific reference for the research and development of Guizhou medicinal resource by screening and evaluation the RAW264.7 cell activity of 81 kinds of extracts from medicinal plants. Methods RAW264.7 cells were treated by samples along or with Lipopolysaccharide (LPS) for some time, the expression of TNF-ct in supernatant was analyzed by ELISA to evaluate the effect of samples on the RAW264.7 cells activation. Result 34 samples up-regulated the expression of TNF-α, and 2 samples inhibited the expression of TNF-α induced by LPS. Conclusion Some extracts can stimulate the activity of RAW264.7 cell; some extracts can inhibit the activity of RAW264.7 cell induced by LPS. This work provides substructure and reference for the research and development.
